锥栗MADS-box基因家族鉴定及组织特异性表达分析 被引量:3

摘要 为研究MADS-box基因家族在锥栗花和果实发育过程中的作用,在锥栗全基因组水平上,用生物信息学方法对其MADS-box基因家族进行鉴定、理化性质分析、系统进化分析和染色体定位,分析其基因复制、基因结构、顺式作用元件和表达量。结果表明,锥栗基因组中存在60个ChM ADS-box基因,不均匀地分布在除了2号染色体(Chr2)外的11条染色体上,其中,存在4对串联重复和8对片段重复。根据系统发育关系,可将锥栗基因组的60个ChM ADS-box基因分为type-Ⅰ型(20个)和type-Ⅱ型(40个)。ChM ADS-box蛋白均含有MADS结构域(Motif1和Motif2),且同亚家族基因显示出相似的基因结构。ChM ADS-box基因的启动子区域含有植物生长、光反应、激素反应和胁迫反应等顺式作用元件。ChM ADS-box基因在叶、雄花、雌花、果实和壳斗5种组织中具有特异性表达模式,表明其在植物生长发育过程中具有不同的功能。在果实和雄花发育的3个过程中,ChM ADS17、ChM ADS22、ChM ADS30、ChM ADS32、ChM ADS36、ChM ADS43、ChM ADS49、ChM ADS53表达较高,表明它们在锥栗花和果实发育过程中起到重要作用。 To study the role of the MADS-box gene family in the flower and fruit development of Castanea henryi, bioinformatics methods were used to identify MADS-box gene family members and their physical and chemical properties, chromosomal location, gene replication, gene structure, cis-acting elements, and transcriptional expression at the genome-wide level. We identified 60 ChMADS-box genes in the C. henryi genome. The genes were unevenly distributed on 11 chromosomes, with none on chromosome 2(Chr2). There were four pairs of tandem repeats and eight pairs of fragment repeats in these genes. Based on the phylogenetic relationship, the genes were divided into type-Ⅰ(20) and type-Ⅱ(40). All ChMADS-box proteins contained MADS domains(Motif1 and Motif2) and showed similar gene structures with subfamily genes. The promoter region of ChMADS-box genes contained cis-acting elements, such as plant growth, light response, hormone response, and stress response elements. ChMADS-box genes showed specific expression patterns in the leaf, cupule, male flower, female flower, and fruit, indicating that they have different functions in plant growth and development. ChMADS17, ChMADS22, ChMADS30, ChMADS32, ChMADS36, ChMADS43, ChMADS49, and ChMADS53 were highly expressed in the three stages of fruit and male flower development, indicating that they play an important role in flower and fruit development in C. henryi.
