摘要
【目的】旨在研究持续禁食与恢复采食状态下食欲调控中枢神经元兴奋性的变化,挖掘调控动物采食的潜在神经核团。【方法】以7周龄C57BL/6雄性小鼠为试验对象,采用c-Fos免疫荧光技术检测不同核团在持续禁食(饥饿38 h)和恢复采食(饥饿36 h+恢复采食2 h)状态下的兴奋性变化,通过数据分析筛选出参与食欲调控的潜在中枢核团。【结果】摄食之后,前缘皮层(PrL)、外侧隔核(LS)、前背视前核(ADP)、弓状核(ARC)、水管周灰质(PAG)、未定带(ZI)、外侧缰核(LHb)、下丘脑背内侧核(DMH)、下丘脑后区(PH)和侧视前区(LPO)的c-Fos表达显著下降(P<0.05);室旁核(PVN)、视上核(SO)、下丘脑腹内侧核(VMH)和孤束核(Sol)的c-Fos表达显著升高(P<0.05)。【结论】研究进一步证实中枢神经系统中弓状核(ARC)、室旁核(PVN)等已知核团参与了动物的食欲调控,同时发现:前缘皮层(PrL)、前背视前核(ADP)和下丘脑后区(PH)等核团也可能在动物食欲调控中发挥重要作用。
[Objective]The aim of this study was to investigate the changes of neuronal excitability in the appetite regulatory center under fasting and refeeding,thus identifying the potential nuclei which regulate feeding.[Method]C57BL/6 male mice aged 7 weeks were selected as the experimental subjects.C-fos immunofluorescence technique was used to detect the excitability changes of different nuclear groups under fasting(38 h of fasting)and refeeding(36 h of fasting+2 h of refeed).The potential central nuclear groups involved in appetite regulation were screened by data analysis.[Results]After feeding,the expressions of prelimbic cortex(PrL),lateral septal nucleus(LS),anterodorsal preoptic nucleus(ADP),arcuate hypothalamic nucleus(ARC),periaqueductal gray(PAG),zona incerta(ZI),lateral habenular nucleus(LHb),dorsomedial hypothalamus nucleus(DMH),posterior hypothalamic area(PH),and lateral preoptic area(LPO)were significantly decreased(P<0.05).C-Fos expression was significantly increased in paraventricular hypothalamic nucleus(PVN),supraoptic nucleus(SO),ventromedial hypothalamic nucleus(VMH)and solitary tract nucleus(Sol)(P<0.05).[Conclusion]This study proves that,in addition to the arcuate nucleus and paraventricular nucleus,the functional nuclei such as prelimbic cortex(PrL),anterior dorsal preoptic nucleus(ADP)and posterior hypothalamus(PH)may also play important roles in appetite regulation in central nervous system.
