火针调控白细胞介素23/辅助性T细胞17轴干预银屑病炎性机制的研究 被引量:1

摘要 目的观察火针对咪喹莫特诱导小鼠皮肤银屑病样炎性反应的作用,探究火针干预银屑病的可能机制。方法选取18只BALB/c雄性小鼠随机分为对照组、模型组和火针组,每组6只。模型组和火针组小鼠每日背部皮肤涂抹5%咪喹莫特乳膏,连续9天制备银屑病模型,对照组涂抹等量凡士林。火针组在第4到第9天每日给予双侧内关、双侧足三里火针点刺治疗。通过苏木精—伊红染色观察皮肤组织病理变化,酶联免疫吸附测定法检测小鼠血清白细胞介素23、白细胞介素17、白细胞介素10表达水平。结果与对照组相比,模型组小鼠血清白细胞介素10表达水平显著下降(P<0.05),白细胞介素23和白细胞介素17表达水平显著上升(P<0.05);与模型组相比,火针组小鼠血清白细胞介素10表达水平显著上升(P<0.05),白细胞介素23和白细胞介素17表达水平显著下降(P<0.05)。结论火针可能通过促进白细胞介素10表达来抑制白细胞介素23/辅助性T细胞17轴,从而发挥抗炎作用以减轻银屑病样皮损病变。 Objective To observe the effects of fire acupuncture on inflammatory response in mice with psoriasis-like skin induced by imiquimod,and to explore the possible mechanism of fire acupuncture in psoriasis.Methods 18 BALB/c male mice were randomly divided into the control group,the model group and the fire needling group,6 mice in each group.The mice in the model group and the fire needling group were smeared with 5%imiquimod cream on the back for 9 consecutive days to prepare a psoriasis-like skin lesions,while the mice in the control group was smeared with the same amount of Vaseline.The mice in the fire needling group was given fire-needle acupuncture at bilateral Zusanli acupoint(ST36)and bilateral Neiguan acupoint(PC6)every day from the 4th day to the 9th day.The skin tissue was stained with hematoxylin-eosin to observed pathological changes,and the expression levels of interleukin-23,interleukin-17 and interleukin-10 in mice serum were detected by enzyme-linked immunosorbent assay.Results Compared with the mice in the control group,the expression level of interleukin-10 in the model group was significantly decreased(P<0.05),while interleukin-23 and interleukin-17 were significantly increased(P<0.05).Compared with the mice in the model group,the expression levels of interleukin-10 in fire needling group was significantly increased(P<0.05),while interleukin-23 and interleukin-17 were significantly decreased(P<0.05).Conclusion Fire acupuncture may inhibit the interleukin-23/T help cell 17 axis by promoting the expression of interleukin-10,thereby exerting an anti-inflammatory effect to reduce psoriasis-like skin lesions.
