TUFT1在直肠癌组织中表达及其临床诊断和预后价值

The expression of TUFT1 in rectal cancer and its clinical diagnosis and prognostic value

目的探究釉丛蛋白1(TUFT1)在直肠癌组织中的表达水平及其对直肠癌的诊断和预后价值。方法利用Ualcan数据库分析TUFT1 mRNA在正常直肠组织及直肠癌组织中的表达水平;选取2014年12月~2018年4月新乡市第一人民医院诊治的77例直肠癌患者为研究对象,实时荧光定量PCR(qRT-PCR)法测定直肠癌组织、癌旁正常组织TUFT1 mRNA表达水平;分析直肠癌组织TUFT1 mRNA表达水平与直肠癌患者临床病理特征的关系;受试者工作特征曲线(ROC曲线)评价直肠组织TUFT1 mRNA表达水平对直肠癌的诊断价值;Kaplan-Meier生存曲线分析直肠癌组织TUFT1 mRNA表达水平与直肠癌患者预后的关系;COX回归分析影响直肠癌患者预后的因素。结果Ualcan数据库分析显示,直肠癌组织TUFT1 mRNA表达水平高于正常直肠组织(P<0.05);qRT-PCR法检测显示,直肠癌组织TUFT1 mRNA表达水平高于癌旁正常组织(P<0.05);直肠癌组织TUFT1 mRNA表达水平与直肠癌患者淋巴结转移、TNM分期、分化程度相关(P<0.05);直肠组织TUFT1 mRNA表达水平诊断直肠癌的曲线下面积(AUC)为0.889,其截断值为1.59,此时敏感度为83.1%,特异性为87.0%;TUFT1 mRNA低表达组直肠癌患者总生存率高于TUFT1 mRNA高表达组(P<0.05);淋巴结转移、TNM分期、TUFT1 mRNA是影响直肠癌不良预后的独立危险因素(P<0.05)。结论直肠癌患者癌组织中TUFT1 mRNA表达水平较高,均与淋巴结转移、TNM分期、分化程度及预后相关,TUFT1 mRNA可作为诊断直肠癌、评估患者预后的潜在指标。

Objective To explore the expression level of Tuftelin 1(TUFT1)in rectal cancer and its diagnostic and prognostic value for rectal cancer.Methods Ualcan database was used to analyze the expression level of TUFT1 mRNA in normal rectal tissues and rectal cancer tissues;77 patients with rectal cancer diagnosed and treated in our hospital from December 2014 to April 2018 were selected as the research objects;the expression level of TUFT1 mRNA in rectal cancer tissues and normal adjacent tissues was determined by real-time fluorescent quantitative PCR(qRT-PCR)method;the relationship between the expression level of TUFT1 mRNA in rectal cancer tissue and the clinicopathological characteristics of patients with rectal cancer was analyzed;the diagnostic value of rectal tissue TUFT1 mRNA expression level for rectal cancer was evaluated by receiver operating characteristic curve(ROC curve);the relationship between the expression of TUFT1 mRNA in rectal cancer tissue and the prognosis of patients with rectal cancer was analyzed by Kaplan-Meier survival curve;the factors affecting the prognosis of patients with rectal cancer were analyzed by COX regression.Results Ualcan database analysis showed that the expression level of TUFT1 mRNA in rectal cancer tissues was higher than that in normal rectal tissues(P<0.05);the qRT-PCR method showed that the expression level of TUFT1 mRNA in rectal cancer tissue was higher than that in normal adjacent tissues(P<0.05);the expression level of TUFT1 mRNA in rectal cancer was correlated with lymph node metastasis,TNM stage and differentiation degree(P<0.05);the area under the curve(AUC)of rectal tissue TUFT1 mRNA expression level for diagnosis of rectal cancer was 0.889,and its cut-off value was 1.59.At this time,the sensitivity was 83.1%and the specificity was 87.0%;the overall survival rate of rectal cancer patients in the TUFT1 mRNA low expression group was higher than that in the TUFT1 mRNA high expression group(P<0.05);lymph node metastasis,TNM staging,and TUFT1 mRNA were independent risk factors that affected the poor prognosis of rectal cancer(P<0.05).Conclusion The expression level of TUFT1 mRNA in cancer tissues of patients with rectal cancer is relatively high,which is related to lymph node metastasis,TNM staging,degree of differentiation and prognosis.TUFT1 mRNA can be used as a potential indicator for diagnosing rectal cancer and assessing the prognosis of patients.
