超声辅助双水相体系提取枳壳总黄酮及其抗氧化活性研究 被引量:1

Extraction of Total Flavonoids from Fructus Aurantii by Ultrasonic-assisted Aqueous Two-phase System and Its Antioxidant Activity

摘要 采用超声辅助丙酮/NaH_(2)PO_(4)·2H_(2)O双水相体系提取枳壳总黄酮,通过单因素实验和正交实验优化提取工艺,并通过DPPH法和ABTS法评价枳壳总黄酮的体外抗氧化活性。结果表明,枳壳总黄酮的最佳提取工艺为:液料比60∶1(g∶g)、提取时间50 min、提取温度70℃、NaH_(2)PO_(4)·2H_(2)O质量分数23%、丙酮质量分数45%,在此条件下,枳壳总黄酮得率为75.90 mg·g^(-1);枳壳总黄酮具有一定的抗氧化活性,浓度为800μg·mL^(-1)时,其对DPPH自由基的清除率为98.57%,IC_(50)值为48.96μg·mL^(-1);浓度为3000μg·mL^(-1)时,其对ABTS自由基的清除率为98.40%,IC_(50)值为207.71μg·mL^(-1)。 We extracted the total flavonoids from Fructus Aurantii by an ultrasonic-assisted acetone/sodium dihydrogen phosphate dihydrate(NaH2PO4·2 H2O)aqueous two-phase system.Moreover,we optimized the extraction process of total flavonoids from Fructus Aurantii by single-factor experiments and orthogonal experiments,and evaluated the antioxidant activity of total flavonoids from Fructus Aurantii by DPPH method and ABTS method.The results show that the optimal extraction conditions can be determined as follows:the liquid-solid ratio of 60∶1(g∶g),the extraction time of 50 min,the extraction temperature of 70℃,the mass fraction of NaH2PO4·2 H2O of 23%,and the mass fraction of acetone of 45%.Under above conditions,the yield of total flavonoids from Fructus Aurantii can reach 75.90 mg·g^(-1).When the concentration of total flavonoids from Fructus Aurantii is 800μg·mL^(-1),the scavenging rate of DPPH free radicals is 98.57%,and the IC_(50)value is 48.96μg·mL^(-1);when the concentration is 3000μg·mL^(-1),the scavenging rate of ABTS free radicals is 98.40%,and the IC_(50)value is 207.71μg·mL^(-1),indicating that the total flavonoids from Fructus Aurantii has certain antioxidant activity.
