摘要
研究了反相聚合物填料层析纯化万古霉素工艺。得到了较佳工艺条件为:载样量3.5%,洗杂、洗脱流速2.25 BV·h^(-1),先用3.5 BV的6%甲醇-水溶液洗杂,再用12%甲醇-水溶液洗脱。层析纯化后的万古霉素纯度由原来的93.20%提高到98.22%以上,收率达81%以上。该层析工艺降低杂质效果明显,可在较高的效率下获得高纯度万古霉素,有较高的应用价值。
We studied the purification process of Vancomycin by reversed-phase polymer packing chromatography.The optimal process conditions can be determined as follows: the loading amount is 3.5%,the flow rate of impurity-washing and elution is 2.25 BV·h^(-1),the 3.5 BV 6% methanol-aqueous solution is adopted to wash impurity, and 12% methanol-aqueous solution is adopted to elution.After purification by chromatography, the purity of Vancomycin can increase from 93.20% to 98.22%,and the yield can reach above 81%.The chromatography process is effective in reducing impurities, and can obtain high purity Vancomycin with high efficiency, which has a good application value.
