摘要
目的探讨蝙蝠葛碱对恶性胶质瘤细胞增殖和凋亡的影响及机制。方法以人恶性胶质瘤细胞株U87为研究对象,采用MTT法检测不同浓度蝙蝠葛碱在48h内对U87细胞增殖的作用;平板克隆形成实验检测蝙蝠葛碱对U87细胞克隆形成能力的影响;Hoechst染色法检测不同浓度蝙蝠葛碱对U87细胞凋亡的作用;Western blot检测蝙蝠葛碱对凋亡相关蛋白p53、Bax、Bcl-2、Caspase-3和PARP表达的影响。结果与对照组相比,蝙蝠葛碱可以浓度依赖性地抑制U87细胞增殖,促进细胞凋亡;增加p53、Bax、Caspase-3和PARP蛋白并下调Bcl-2蛋白表达。结论蝙蝠葛碱对神经胶质瘤的增殖抑制作用可能是通过调控凋亡相关蛋白实现。
Objective To investigate the effects of Dauricine, a natural traditional Chinese medicine monomer small molecule compound, on the proliferation and apoptosis of malignant glioma cells and the related mechanism. Methods The human malignant glioma cell line U87 was used as the research object.MTT assay was used to detect the proliferation of U87 at different concentrations of Dauricine at 48 h.The effect of dauricine on the colony formation ability of U87 cells was detected by plate cloning assay.The effect of Dauricine on the apoptosis of U87 cells was detected by Hoechst staining.The effect of Dauricine on the expression of apoptosis-related proteins like p53,Bax, Bcl-2,Caspase-3 and PARP(poly ADP-ribose polymerase, PARP) was detected by Western blot.Results Compared with the control group, Dauricine dose-dependently inhibited the proliferation of U87 cells and promoted cell apoptosis.Dauricine dose-dependently increased the protein levels of p53,Bax, Caspase-3 and PARP, and decreased the expression of Bcl-2.Conclusion Dauricine may exert its inhibitory effects on glioblastoma cell proliferation through regulating the expression of apoptosis-related proteins.
