基于生物信息学分析胰腺癌肿瘤浸润性白细胞组成和预后相关的免疫基因

Analysis of tumor-infiltrating leukocyte compositions and prognostic immunerelated genes in pancreatic cancer based on bioinformatics

摘要 目的:鉴定胰腺癌(PC)浸润性白细胞组成和预后相关的免疫相关基因(IRGs),为患者预后预测提供依据。方法:根据TCGA和GTEx数据库,研究PC和正常胰腺组织中白细胞组成的差异,并分析178例PC患者中差异表达的与OS相关的IRGs。利用计算生物学的方法分析这些IRG的潜在作用机制和性质。采用多变量Cox分析探寻新的PC预后生物标志物。结果:与非肿瘤组织相比,PC组织中CD4^(+)记忆性T细胞和M0巨噬细胞更为常见且占优势。功能富集分析表明,OS相关的差异表达IRGs积极参与了PI3K-Akt信号通路。12个预后相关差异表达的IRGs可用于预后预测。结论:筛选出12个PC特异性IRGs,可作为预测预后的新指标。 Objective:To identify the immune-related genes(IRGs)related to the composition of infiltrating leukocytes and prognosis in pancreatic cancer(PC),and to provide basis for predicting the prognosis of patients with PC.Methods:According to TCGA and GTEx database,the difference of leukocyte composition between PC and normal pancreatic tissue was studied,and the differentially expressed IRGs related to OS in 178 patients with PC were analyzed.The potential mechanism and properties of these IRGs were analyzed by means of computational biology.Multivariate Cox analysis was used to explore new biomarkers of PC prognosis.Results:Compared with non-tumor tissues,CD4^(+)memory T cells and M0 macrophages were more common and dominant in PC tissues.Functional enrichment analysis showed that OS-related differentially expressed IRGs was actively involved in PI3K-Akt signaling pathway.Twelve differentially expressed IRGs related to prognosis could be used to predict prognosis.Conclusion:Twelve PC-specific IRGs have been screened,which can be used as a new index to predict the prognosis.
