摘要
目的:基于液相质谱联用(LC-MS)测定中药复方半夏泻心汤大鼠含药血清及含药肠吸收液中4种特征成分含量从而建立实验用药质量标准。方法:利用LC-MS技术对半夏泻心汤大鼠含药血清及含药肠吸收液中4种特征成分进行定量分析,并进行方法学考察。结果:半夏泻心汤含药肠吸收液及含药血清中均测得黄芩苷、人参皂苷Rb1、小檗碱、甘草酸。含药肠吸收液及含药血清中含量最高成分均为黄芩苷,含量分别为(115.30±5.81)μg/m L、(52.69±1.21)ng/mL;含药肠吸收液中含量最低成分为甘草酸,含量为(5.35±0.08)ng/mL。含药血清中含量最低成分为小檗碱,含量为(1.80±0.12)ng/mL。结论:该方法可以快速、准确的检测半夏泻心汤大鼠肠吸收液、血清中特征成分甘草酸、小檗碱、人参皂苷Rb1、黄芩苷的含量差异,为后续药理实验中半夏泻心汤应用提供质量控制和理论依据。
Objective:To determine four characteristic components in rat serum and intestinal absorption solution of Banxia Xiexin Decoction by liquid chromatography-mass spectrometry(LC-MS),the experimental drug quality standard was established.Methods:Four characteristic components in drug containing serum and drug containing intestinal absorption solution of Banxia Xiexin Decoction rats were quantitatively analyzed by LC-MS,and the methodology was investigated.Results:Baicalin,ginsenoside Rb1,berberine and glycyrrhizic acid were measured in the intestinal absorption solution and serum of Banxia Xiexin Decoction.Baicalin was the highest component in the drug containing intestinal absorption solution and drug containing serum,with the content of(115.30±5.81)μg/mL,(52.69±1.21)ng/mL,respectively.Glycyrrhizic acid was the lowest component in the drug containing intestinal absorption solution,with a content of(5.35±0.08)ng/mL.Berberine was the lowest component in the serum containing medicine,and the content was(1.80±0.12)ng/m L.Conclusion:This method can quickly and accurately detect the content differences of characteristic components glycyrrhizic acid,berberine,ginsenoside Rb1 and baicalin in intestinal absorption solution and serum of Banxia Xiexin Decoction rats,and provide quality control and theoretical basis for the application of Banxia Xiexin Decoction in subsequent pharmacological experiments.
