Study on the difference of chemical constituents of Paeonia veitchii Lynch in different harvesting periods based on UHPLC-Q-Exactive Orbitrap MS

摘要 目的:建立超高效液相色谱四极杆-静电场轨道阱高分辨质谱(UHPLC-Q-Exactive Orbitrap MS)分析壤塘地区不同采收期川赤芍成分差异的方法,为科学评价壤塘地区开发种植川赤芍及其适宜采收期提供科学依据。方法:采用Waters Acquity UHPLC BEH C_(18)色谱柱(2.1 mm×50 mm,1.7μm),以乙腈(A)-0.1%甲酸水溶液(B)为流动相梯度洗脱。在负离子模式下采集MS数据,依据质谱谱图信息分析赤芍药材的化学成分。运用统计学方法筛选壤塘地区川赤芍和道地产区川赤芍的指标性差异成分,比较其相对含量。结果:从壤塘川赤芍中共鉴定出包含单萜、黄酮等37种化学成分,指标性成分含量比较显示壤塘地区适宜川赤芍的种植且其最佳采收期在10月份左右。结论:本研究所建方法可用于壤塘川赤芍中有效成分的快速分析,并为壤塘地区作为开发种植川赤芍的区域之一及确定合理采收期提供了科学依据。 Objective:To establish a method of ultra high performance liquid chromatography(UHPLC-Q-Exactive Orbitrap MS)to analyze the differences of chemical components of Paeonia veitchii Lynch in different harvest periods in Rangtang area of Sichuan Province,so as to provide a scientific evaluation about the suitable harvest time of Paeonia veitchii Lynch in Rangtang area.Methods:The Waters Acquity UHPLC BEH C_(18)chromatographic column(2.1 mm×50 mm,1.7μm)was gradient eluted with acetonitrile(A)-0.1%formic acid aqueous solution(B)as mobile phase.MS data were collected in negative ion mode,and the chemical components of Paeonia veitchii Lynch were analyzed according to the mass spectrum information.Statistical methods were used to screen the index difference components between Paeonia veitchii Lynch in Rangtang area and Paeonia veitchii Lynch in genuine production area,and compare their relative contents.Results:A total of 37 chemical components including monoterpenes and flavonoids were identified from Paeonia veitchii Lynch in Rangtang.The comparison of index components showed that Rangtang was suitable for planting Paeonia veitchii Lynch and the best harvest time was around October.Conclusion:The method established in this study can be used for rapid analysis of effective components of Paeonia veitchii Lynch in Rangtang area,and provide scientific basis for Rangtang area as one of the areas for developing and planting Paeonia veitchii Lynch and determining the reasonable harvest time.
