针刺十三鬼穴对抑郁模型大鼠脑组织中kir4.1蛋白表达的影响 被引量:3

Effects of acupuncture at thirteen ghost points on the expression of kir4.1 protein in the brain tissue of depression model rats

摘要 目的:探究针刺上星、大陵穴对慢性不可预测轻度应激(CUMS)抑郁模型大鼠外侧缰核、前额皮质、海马中kir4.1蛋白表达的影响。方法:48只SD大鼠随机分为对照组、模型组、针刺组、氟西汀组,每组12只。采用CUMS结合孤养建立抑郁模型,针刺组于造模前1 h给予针刺干预,隔日1次,每次20 min,共计11次。氟西汀组于造模前1 h给予氟西汀(2.1 mg/kg)灌胃干预,1次/d,共计21 d。通过体质量、糖水偏好试验、强迫游泳实验观察大鼠抑郁样行为表现;Western Blot检测大鼠外侧缰核、前额皮质、海马中kir4.1蛋白表达情况。结果:造模后,与对照组比较,模型组体质量显著下降(P<0.01)。与模型组比较,针刺组和氟西汀组体质量显著提升(P<0.01),针刺和氟西汀治疗后大鼠糖水偏好指数显著增加(P<0.01),不动时间显著减少(P<0.05,P<0.01)。与对照组比较,模型组大鼠外侧缰核、前额皮质和海马中kir4.1蛋白表达均显著增加(P<0.01);针刺组能逆转大鼠前额皮质中kir4.1蛋白表达(P<0.01),氟西汀组能降低kir4.1在外侧缰核、海马和前额皮质的表达(P<0.05,P<0.01)。结论:针刺十三鬼穴通过降低大鼠脑中kir4.1蛋白表达,起到预防大鼠发生抑郁样行为效果,这可能是针刺抗抑郁的潜在作用机制之一。 Objective:To explore the effect of acupuncture at the Shangxing(GV 23)and Daling(PC 7)points on the expression of kir4.1 protein in the lateral habenula,prefrontal cortex and hippocampus in depression model rats induced by chronic unpredictable mild stress(CUMS).Methods:A total of 48 SD rats were randomly divided into normal group,model group,acupuncture group and fluoxetine group,with 12 rats in each group.To create depression-like behavior in groups except for the normal one,rats were exposed to CUMS combined with isolation.The acupuncture group was given acupuncture intervention 1 hour before modeling,once every other day,20 mins per time,for a total of 11 times.Rats in fluoxetine group,fluoxetine(2.1 mg/kg)was given intragastric intervention every day 1 hour before modeling,for a total of 21 days.Depression-like behaviors of rats was observed with body weight,sucrose preference test and forced swimming test.Western Blot was used to detect kir4.1protein expression in rats’lateral habenula,prefrontal cortex and hippocampus.Results:After modeling,compared with the control group,the weight of the model group was significantly decreased(P<0.01).Compared with the model group,the weight of the acupuncture group and fluoxetine group was significantly increased(P<0.01).Behavioral studies showed that after acupuncture and fluoxetine treatment,the sucrose preference rate increased significantly(P<0.01),and the immobility time was significantly reduced(P<0.05,P<0.01).Compared with the control group,the expression of kir4.1 protein in the lateral habenula,prefrontal cortex and hippocampus of the model group increased significantly(P<0.01).After acupuncture treatment,the expression of kir4.1protein in the prefrontal cortex of rats could be reversed(P<0.01).Fluoxetine treatment could also reduce the expression of kir4.1in the lateral habenula,hippocampus and prefrontal cortex(P<0.05,P<0.01).Conclusion:Acupuncture at thirteen ghost points can prevent depression-like behaviors in rats by reducing the expression of kir4.1 in the brain of rats.This may be one of the potential antidepressant mechanisms of acupuncture.
