补肾解毒通络方对急性淋巴细胞白血病骨髓微环境间充质干细胞修复作用 被引量:1

Repair effect of mesenchymal stem cells from bone marrow microenvironment of acute lymphoblastic leukemia by Bushen Jiedu Tongluo Formula

摘要 目的:研究补肾解毒通络方对急性淋巴细胞白血病(ALL)骨髓微环境的间充质干细胞(MSCs)的修复作用。方法:将BALB/C小鼠随机分为正常对照组、模型对照组、西药治疗组、补肾解毒通络组。通过照射后尾静脉注射L1210细胞株制备ALL模型。采用镜下观察骨髓涂片评估造模及治疗效果,集落培养检测细胞增殖,流式细胞术检测凋亡,CCK-8检测细胞存活率,ELISA检测小鼠骨髓微环境MSCs相关细胞因子(SCF、CCR4、CXCL12、ICAM1)的分泌。结果:骨髓涂片显示造模成功,补肾解毒通络组小鼠骨髓MSCs的细胞集落数较西药治疗组显著增多(P<0.05),而细胞凋亡率较西药组显著下降(P<0.05);补肾解毒通络组和西药治疗组的细胞活性均显著高于模型对照组(P<0.01),ELISA结果显示,补肾解毒通络方对SCF、CCR4、CXCL12、ICAM1的分泌均有影响,且在MSCs与CD_(34)^(+)细胞共培养时效果更好。结论:补肾解毒通络方可以修复ALL小鼠骨髓微环境的MSCs。 Objective:To study the repair effect of Bushen Jiedu Tongluo Formula on mesenchymal stem cells (MSCs)in acute lymphoblastic leukemia (ALL) bone marrow microenvironment.Methods:BALB/C mice were randomly divided into normal control group,model control group,medicine treatment group,Bushen Jiedu Tongluo group.ALL model was established by injecting L1210 cell line into caudal vein after irradiation.Bone marrow smear was observed under microscope to evaluate the modeling and therapeutic effect,cell proliferation was detected by colony culture,apoptosis was detected by flow cytometry,cell survival rate was detected by CCK-8,and secretion of cytokines related to mouse bone marrow microenvironment was detected by ELISA.Results:The bone marrow smear showed that the model was successfully established,the cell colony of bone marrow mesenchymal stem cells in the Bushen Jiedu Tongluo group were significantly increased than that in the medicine treatment group(P<0.05),while the apoptosis in Bushen Jiedu Tongluo group significantly decreased than that in the medicine treatment group(P<0.05),the cell activity of the Bushen Jiedu Tongluo group and the medicine treatment group were significantly higher than that of the model control group (P<0.01),ELISA results showed that Bushen Jiedu Tongluo group had effects on the secretion of SCF,CCR4,CXCL12 and ICAM1,and the effect was more significant when the MSCs were co-cultured with CD_(34)^(+) cells.Conclusion:Bushen Jiedu Tongluo Formula can repair MSCs in bone marrow microenvironment of ALL mice.
