摘要
提升鲜食玉米叶酸含量是营养强化育种的重要课题之一。为高效创制高叶酸鲜食玉米新种质,本研究根据叶酸代谢关键基因ZmGFT1的目标变异位点,设计基于碱煮法DNA提取的3对PARMS标记并对应用效果进行评估。结果表明,三对引物识别基因型与测序结果一致,引物2无法识别杂合基因型,引物1识别率高、针对性强、效果最好;通过PARMS识别到的GGG基因型材料平均叶酸含量显著高于GAA、CAA两种基因型。该方法为未来鲜食玉米营养强化育种及高通量分子标记辅助选择育种提供实用手段。
Improving the folic acid content of fresh corn is one of the important topics of nutrition intensification breeding. In order to efficiently create new high-folate fresh corn germplasm, according to the target variant loci of the key gene ZmGFT1 in folic acid metabolism, three PARMS markers based on alkaline boiling DNA extraction were designed and the application effects were evaluated. The results showed that the genotypes identified by primers were consistent with the sequencing results. Primer 2 could not identify heterozygous genotypes. Primer 1 had a high recognition rate, strong pertinence, and the best effect. The average folic acid content of GGG genotype materials identified by PARMS was significantly higher than that of GAA and CAA. This method provides a practical means for future fresh corn nutrition-enhanced breeding and high-throughput molecular marker-assisted selection.
