摘要
目的 通过制备三七有效成分原位凝胶干预大鼠牙周炎模型,验证三七凝胶对牙周炎的疗效。方法 将180 g左右的SD大鼠建立慢性牙周炎模型并分为2组,分别使用制备的三七有效成分原位凝胶和生理盐水注射于炎症处,每2 d干预注射1次持续1周,之后用酶联免疫吸附实验(ELISA)检测观察SD大鼠血清及唾液内前列腺素2(PGE2)、肿瘤坏死因子α(TNF-α)表达情况,并使用小动物CT(Micro-CT)系统观察2组大鼠的牙槽骨吸收情况。结果 药物干预后检测牙周炎模型大鼠唾液及血液PGE2、TNF-α与对照组相比显著降低(P<0.05);CT影像显示,大鼠建模组较正常对照组牙槽骨为垂直吸收,药物干预组较建模组牙槽骨密度增高。结论 三七原位凝胶对牙周炎模型治疗有一定疗效,为三七治疗牙周炎提供理论支持。
Objective To verify the effect of Panax notoginseng on periodontitis in rats by preparing in situ gel(ISG).Methods 180 g SD rats were used to established chronic periodontitis models and divided into two groups. In situ gel of the active components of Panax notoginseng and normal saline were injected into the inflammatory site respectively. The intervention injection was performed once every two days for a week. After that, the enzyme linked immunosorbent assay(ELISA) was used to detect prostaglandin 2(PGE2) and tumor necrosis factor(TNF-α) in the serum and saliva of rats. Micro-CT system was used to observe the alveolar bone resorption in the two groups.Results After drug intervention, PGE2 and TNF-A in saliva and serum of rats with periodontitis were significantly decreased compared with the control group(P<0.05). CT images showed that the alveolar bone in the model group was vertically absorbed compared with the control group,and the alveolar bone density in the drug intervention group was higher than that in the model group. Conclusion Panax notoginseng in situ gel has a certain effect on periodontitis model,which provides theoretical support for the treatment of periodontitis by Panax notoginseng.
