摘要
越来越多的证据表明,Warburg效应与恶性肿瘤的代谢机制有关。这种代谢重编程为肿瘤细胞生长提供优势,为肿瘤细胞转移创造合适环境,并帮助其逃避凋亡。Warburg效应的内在机制十分复杂,可能与癌基因激活、抑癌基因失活、肿瘤微环境改变和糖代谢酶异常表达等有关。酶途径的代谢改变可导致糖酵解能力增强,满足肿瘤细胞主动增殖的能量需求,从而进一步支持肿瘤的生长发展。本文主要从酶学角度回顾和总结了近年来Warburg效应在非小细胞肺癌细胞中的代谢机制。
There is increasing evidence that the Warburg effect may be related to the metabolic mechanisms of malignant tumors.This metabolic reprogramming provides an advantage for tumor cell growth,and creates a suitable environment for tumor cell metastasis,as a support to escape apoptosis.The internal mechanism of Warburg effect is very complicated,which may be related to oncogene activation,tumor suppressor gene inactivation,tumor microenvironment changes and abnormal expression of glucose metabolism enzymes.Changes in the enzyme metabolic can lead to enhanced glycolysis capacity and meet the energy requirements of tumor cell active proliferation,thus further supporting tumor growth and development.This paper reviews and summarizes the metabolic mechanism of Warburg effect in non-small cell lung cancer cells based on the perspective of enzymology.
