醒脑静联合纳洛酮对急性酒精性昏迷大鼠的促醒作用研究 被引量:1

摘要 目的:研究醒脑静联合纳洛酮对急性酒精性昏迷大鼠的促醒作用,为临床合理用药提供实验依据。方法:取健康SD大鼠随机分为5组,即空白组、模型组、醒脑静组(20 m L/kg)、盐酸纳络酮组(0.9 m L/kg)及醒脑静联合纳洛酮用药组(20 m L/kg+0.9 m L/kg)。除空白组外,其余各组采用32%乙醇以17.8 m L/kg剂量腹腔注射,复制急性酒精中毒大鼠昏迷模型,大鼠翻正反射消失后给予上述剂量的药物治疗,并采用RCS(rat coma scale,RCS)方法进行昏迷评分,评价醒脑静联合纳洛酮用药的促醒作用;同法复制急性酒精性昏迷大鼠模型,采用水迷宫实验,记录各组大鼠进入平台的时间、进入次数及目标象限停留时间所占百分比。结果:与模型对照组比较,单独用药与联合用药组大鼠昏迷评分明显增加;与单用纳络酮相比较,联合用药及单用醒脑静的大鼠昏迷评分分别在造模给药后2~4 h与2~4.5 h内明显升高,联合用药组和醒脑静组较单用纳洛酮组评分恢复到清醒所用时间提前了1h(评分≥10),且联合用药组疗效略优于单用醒脑静组。此外,水迷宫实验结果显示,与模型组相比,单独用药组与联合用药组均能减少大鼠逃避潜伏期,增加穿越平台次数以及目标象限停留所占百分比;与纳洛酮组相比较,单用醒脑静组及二者联合用药组大鼠逃避潜伏期明显减少,穿越平台次数及目标象限停留所占百分比明显增加;其中联合用药组在减少逃避潜伏期、增加穿越平台次数及目标象限停留所占百分比优于单用醒脑静。结论:醒脑静联合纳洛酮用药对急性酒精性昏迷大鼠的促醒作用和学习记忆的恢复作用较好,疗效优于醒脑静和纳络酮单独用药,为临床用药提供了实验支持。 Objective Study the wake-up effect of Xingnaojing combined with naloxone on acute alcoholic coma rats,and to provide experimental basis for clinical rational drug use.Methods Healthy SD rats were randomly divided into 5 groups,namely blank group,model group,Xingnaojing group(20 m L/kg),Naloxone hydrochloride group(0.9 m L/kg)and Xingnaojing combined with Naloxoe medication group(20 m L/kg+0.9 m L/kg).Except for the blank group,the other groups were given intraperitoneal injection of 32%ethanol at a dose of 17.8 m L/kg to replicate the acute alcoholism rat coma model.After the righting reflex disappeared,the rats were given the above doses of drug treatment,and the RCS(Rat Coma Scale,RCS)method was used to score the coma to evaluate the wake-up effect of Xingnaojing combined with naloxone;The rat model of acute alcoholic coma was replicated in the same way,and the water maze test was used to record the time of entering the platform,the number of entries,and the percentage of time spent in the target quadrant.Results Compared with the model control group,the coma scores of the rats in the single drug and combined drug groups were significantly increased;Compared with naloxone alone,the coma scores of the rats treated with the combination and Xingnaojing alone increased significantly within 2-4 h and 2-4.5 h after the model administration,respectively,compared with the single-use naloxone group,the recovery time to wakefulness in the combination group and Xingnaojing group was 1 hour earlier(score≥10),and the efficacy of the combination group was slightly better than that of the single-use Xingnaojing group.In addition,the results of the water maze experiment showed that compared with the model group,both the single drug group and the combined drug group could reduce the escape latency of rats,increase the number of crossing platforms and the percentage of staying in the target quadrant;Compared with the naloxone group,the escape latency of the rats in the Xingnaojing group alone and the combination of the two groups was significantly reduced,and the number of crossing the platform and the percentage of staying in the target quadrant were significantly increased;Among them,the combination group was better than the single-use Xingnaojing in reducing the escape latency,increasing the times of crossing the platform and staying in the target quadrant.Conclusion The Xingnaojing combined with naloxone has better wake-up promoting effect and learning and memory recovery effect in acute alcoholic coma rats,and the curative effect is better than Xinnaojing and naloxone alone,which provides experimental support for clinical medication.
