番茄果皮颜色的基因功能标记开发及色差仪参数相关分析 被引量:2

摘要 番茄果皮颜色由已克隆的SIMYB12基因控制,是影响消费者选择的最直观标准和重要的外观品质之一,利用分子功能标记及精准的果皮颜色测定对果皮颜色进行筛选,对加快番茄果皮选育具有指导意义。根据SIMYB12基因起始密码子上游4 kbp处的603 bp片段缺失,共设计可在琼脂糖凝胶上检测的分子功能标记16对。结果显示,共有7对功能标记可用(4对显性标记和3对共显性标记),其中A-10扩增效果最好,扩增片段大小为890bp(有色果皮)和287 bp(无色果皮)。利用30份番茄种质资源对A-10进行验证,结果表明,A-10能够准确地鉴定供试材料是否包含缺失片段的基因型。利用色差仪对不同颜色的番茄果皮进行测定和分析,Ch和Lab测量值均可用于果皮颜色的定量分析,而Ch测量值更加直观和简便。本研究开发的果皮分子功能标记A-10可用于番茄果皮颜色的分子标记辅助选择育种,并且Ch测量值可用于果皮颜色快速和准确的测定。 The pericarp color of tomato is controlled by cloned SIMYB12 gene, which is one of the most intuitive criteria and important appearance qualities influencing consumer intention. Identification of peel color by molecular functional marker and accurate skin color determination is of guidance to accelerate tomato peel color breeding. Based on a 603 bp deletion at 4 kbp upstream of the SIMYB12 gene, 16 pairs of functional markers were designed, and a total of 7 pairs of functional markers were available(4 dominant and 3 co-dominant pairs), among which the A-10 primer was the most effective, with amplified fragment sizes of 890 bp(colored peel) and 287 bp(colorless peel). Using 30 tomato germplasms to validate A-10 primer, the results showed that A-10 was able to accurately identify whether the test material contained gentotypes for the target deletion region, and tomato pericarp of different colors was measured and analyzed by colorimeter. Both Ch and Lab measurements could be used for quantitative analysis of pericarp color, while Ch measurements were more intuitive and simpler. The molecular functional marker A-10 developed in this study can be used for molecular marker-assisted selection breeding of tomato peel color, and the Ch measurements can be used for rapid and accurate determination of peel color.
