摘要
目的 :探索诱导型一氧化氮合酶(iNOS)在胆管癌细胞的侵袭和迁移过程中的相关作用。方法 :构建靶向iNOS基因的siRNA表达载体,借助脂质体稳定转染QBC939细胞(iNOS-siRNA),同时设置转染无干扰质粒的阴性对照组以及未转染的空白对照组。使用Transwell小室法分别检测3组QBC939细胞侵袭的差异,划痕实验分别检测3组QBC939细胞迁移的差异。结果 :与对照组相比,实验组的iNOS蛋白表达水平明显下降,同时检测到干扰组QBC939细胞侵袭以及迁移明显降低。结论 :iNOS基因与胆管癌的侵袭和迁移相关,特异性沉默iNOS基因来下调其表达能明显抑制人胆管癌QBC939细胞的侵袭及迁移。
Objective To explore the role of iNOS in regulating the migration and invasion of Cholangiocarcinoma cells in vitro. Methods A siRNA expressing vector targeting iNOS was constructed and transfected into QBC939 cells via Lipofectamine 2000 to establish stable transfection cell line. The QBC939 cells transfected non-sense sequence and those untransfected were used as control. The invasion of QBC939 cells were determined by transwell assay, and scratch test was used to analyze the capability of migration. Results Compared with the control subjects, the expression level of iNOS protein in the interference group was significantly reduced;after iONS siRNA, the capability of invasion and migration in the iNOS-siRNA group significantly lower than those two control groups. Conclusions iNOS is associated with the invasion and migration of cholangiocarcinoma.Down-regulation of iNOS decreases invasion and migration of QBC939 cells.
