吸水动力学结合现代技术的当归润制过程研究 被引量:1

摘要 目的运用现代技术量化表征当归的润制过程,阐明当归润制传统经验的科学内涵。方法测定不同润制时间当归的吸水率,构建当归吸水动力学曲线并进行非线性拟合;采用低场核磁共振及磁共振成像技术(LF-NMR/MRI)表征当归润制过程中水分相态及分布状态的变化;采用质构仪测定当归润制过程中质地的变化;采用HPLC法分析当归不同润制时间的阿魏酸含量变化。结果当归润制吸水动力学符合一级动力学方程,决定系数(COD)大于98.1%,非线性拟合结果良好。吸水率随润制温度的增加而增加,随个头的增大而减小。LF-NMR/MRI结果表明当归润制过程中水分由外而内渗入,不同相态水分含量发生明显变化,前4 h内自由水由4.02%增加为95.75%,结合水由95.98%减少为4.25%。除弹性外,当归润制时硬度、压缩功循环等物性指标在8 h内均明显下降,8 h后趋于平稳,符合传统“药透水尽”的判断标准。当归醇溶性浸出物和阿魏酸含量在润制过程中未发生明显变化。结论吸水动力学结合磁场核磁共振和物性分析技术可直观量化表征当归的润制过程,为阐明当归润制传统经验的科学内涵提供了依据。 Objective Quantitative analysis was applied to characterize the moistening process of Danggui(Angelica sinensis)and then to elucidate the scientific connotation of the traditional experiences about it.Methods The water absorption kinetic curve of Danggui(Angelica sinensis)and nonlinear fitting was constructed.Low field nuclear magnetic resonance and magnetic resonance imaging(LF-NMR/MRI)were used to characterize the changes of water phase and distribution during the moistening process of Danggui(Angelica sinensis).The texture changes of Danggui(Angelica sinensis)during moistening were measured by texture analyzer.The content of ferulic acid in Danggui(Angelica sinensis)at different moistening times was analyzed by high performance liquid chromatography(HPLC).Results The water absorption kinetics of Danggui(Angelica sinensis)were in accordance with the first-order kinetic equation,and the coefficient of determination(COD)was greater than 98.1%.Moreover,the water absorption rate increased with increasing of temperature,but decreased with the increasing of sample size.LF-NMR/MRI analysis showed that water entered from outside to inside during the moistening of Danggui(Angelica sinensis),and the water content of different phases changed significantly.Within the first 4 hours,the free water increased from 4.02% to 95.75%,and the bound water decreased from 95.98% to 4.25%.Except for the physical properties of elasticity,hardness and compression work cycle of Danggui(Angelica sinensis)decreased significantly within 8 h,and tended to be stable after that,which was consistent with the traditional criterion“herb soaking with exact amount of water”.The contents of alcohol-soluble extract and ferulic acid of Danggui(Angelica sinensis)remained unchanged during the moistening process.Conclusion The water absorption kinetics combined with LF-NMR/MRI and texture analyzer can visually quantify the moistening process of Danggui(Angelica sinensis),which provides a new train of thought for elucidaing the mechanism of the traditional experiences about moisturizing process of Danggui(Angelica sinensis).
