miR-103对鼻咽癌SUNE1细胞生物学行为的影响

摘要 目的 探讨miR-103对鼻咽癌SUNE1细胞增殖、迁移、侵袭和凋亡的影响。方法 采用脂质体转染试剂将miR-103 mimic、miR-103 NC、miR-103 inhibitor瞬时转染至SUNE1细胞,RT-PCR检测miR-103的转染效率,CCK-8实验检测其细胞增殖能力,Transwell实验检测细胞的迁移能力和侵袭能力,流式细胞术检测其细胞凋亡能力,Western blot检测相关蛋白表达水平。结果 与SUNE1组相比,miR-103 mimic组的miR-103表达量升高,miR-103 inhibitor组的miR-103表达量降低;在48 h和72 h时,miR-103 mimic组细胞生长速率均明显升高,miR-103 inhibitor组生长速率明显降低;miR-103 mimic组细胞迁移能力和侵袭能力均显著提升,而miR-103 inhibitor组均显著减弱;在miR-103 mimic组中vimentin蛋白的表达升高、PARP-1蛋白的表达降低,在miR-103 inhibitor组中vimentin和cyclinD1蛋白的表达降低,PARP-1蛋白的表达升高;差异均具有统计学意义(P<0.01)。各组细胞凋亡率差异无统计学意义(P>0.05)。结论 miR-103能够调控鼻咽癌SUNE1细胞的增殖、迁移和侵袭,但不影响细胞的凋亡。 Objective To investigate the effects of miR-103 on proliferation, migration, invasion, and apoptosis of nasopharyngeal carcinoma SUNE1 cells. Methods MiR-103 mimic, miR-103 NC, and miR-103 inhibitor were transiently transfected into SUNE1 cells with liposome transfection reagent. The transfection efficiency of miR-103 was detected by RT-PCR. The cell proliferation was evaluated by CCK-8 assay. Transwell assay was used to detect the migration and invasiveness of cells.Flow cytometry was used to detect the ability of apoptosis. Western blot was used to detect the expression levels of related proteins. Results Compared with SUNE1 group, the expression of miR-103 in miR-103 mimic group increased, while that in miR-103 inhibitor group decreased. At 48 h and 72 h, the cell growth rate of miR-103 mimic group was significantly increased, while that of miR-103 inhibitor group was significantly decreased. In miR-103 mimic group, the ability of cell migration and invasion were significantly improved, while that in miR-103 inhibitor group were significantly decreased. In miR-103 mimic group, the expression of vimentin protein was increased, and the expression of PARP-1 protein was decreased. In miR-103 inhibitor group, the expressions of vimentin and cyclinD1 protein were decreased, and the expression of PARP-1 protein was increased. The differences were statistically significant(P<0.01). Conclusions MiR-103 can regulate the proliferation, migration, and invasion of SUNE1 cells, but does not affect cell apoptosis.
