摘要
目的:研究针刺对血管性痴呆(VD)大鼠海马CA1区脑红蛋白(Ngb)和低氧诱导因子-1α(HIF-1α)表达的影响。方法:随机分为假手术组、VD模型组以及电针治疗组,选用改良双侧CCA阻断法(2-VO)法建立VD大鼠模型,分别于造模后第3天和针刺治疗结束后,使用Morris水迷宫检测大鼠认知水平,Tunel法观察海马细胞形态,并检测各组海马中的ATP浓度,Western blot法检测Ngb和HIF-1α表达水平,RT-PCR法检测p53RFPmRNA、NgbmRNA、HIF-1αmRNA表达水平。结果:与假手术组比较,造模后大鼠的学习记忆功能受损,海马神经细胞凋亡增加,Ngb水平应激性升高(P<0.05),HIF-1α表达水平增高(P<0.05);较模型组,针刺组大鼠的学习记忆能力及神经元凋亡情况显著改善,ATP浓度增加,p53RFPmRNA下将,Ngb和HIF-1α表达水平显著增高(P<0.05)。结论:针刺可以通过增加海马区Ngb和HIF-1α的表达水平,抑制海马神经元的凋亡,从而改善血管痴呆模型动物学习记忆。
Objective: To study the effect of acupuncture on the expressions of neucleoglobin(Ngb) and hypoxiainducible factor-1α(HIF-1α) in hippocampal CA1 region of rats with vascular dementia(VD). Methods: They were randomly divided into sham operation group, VD model group and electroacupuncture treatment group. The modified bilateral CCA block method(2-VO) method was used to establish the VD rat model, and the rats were treated with acupuncture on the 3rd day after model establishment. After the end, the Morris water maze was used to detect the cognitive level of the rats, the morphology of the hippocampal cells was observed by the Tunel method, and the ATP concentration in the hippocampus of each group was detected, the expression levels of Ngb and HIF-1α were detected by the Western blot method, and the p53RFPmRNA, NgbmRNA, HIF-1αmRNA expression levels. Results: Compared with the sham operation group, the learning and memory function of the rats after modeling was impaired, the apoptosis of hippocampal nerve cells was increased, the level of Ngb was increased(P<0.05), and the expression level of HIF-1α was increased(P<0.05). Compared with the model group, the learning and memory ability and neuron apoptosis of the rats in the acupuncture group were significantly improved, the concentration of ATP was increased, and the expression levels of p53RFP mRNA, Ngb and HIF-1α were significantly increased(P<0.05). Conclusion: Acupuncture can increase the expression levels of Ngb and HIF-1α in the hippocampus and inhibit the apoptosis of hippocampal neurons, thereby improving learning and memory in vascular dementia model animals.
