血清miR-21、miR-150的表达与心房颤动患者NT-proBNP水平的关系及治疗前后变化分析 被引量:1

The correlations between the expression of serum miR-21 and miR150 in patients with atrial fibrillation and the level of NT-proBNP and their changes after treatment

摘要 目的探讨血清微小RNA(miR-21、miR-150)水平与心房颤动和N末端脑钠肽前体(NT-proBNP)水平的相关性,以及心房颤动射频消融术后的变化。方法选取心内科入组心房颤动患者103例(房颤组),入组同期健康者50例为对照组。实时荧光定量PCR技术检测两组的血清中微小RNA-21(miR-21)、微小RNA-150(miR-150)表达。同时检测两组患者血浆NT-proBNP水平。明确miR-21、miR-150与心房颤动和NT-proBNP水平的相关性。同时分析房颤组患者射频消融术后的miR-21、miR-150表达变化及NT-proBNP水平改变。结果miR-21、NT-proBNP水平房颤组显著高于对照组,miR-150水平房颤组显著低于对照组(P<0.05)。miR-21、NT-proBNP水平在持续性心房颤动患者显著高于阵发性心房颤动患者,miR-150水平显著低于阵发性心房颤动患者(P<0.05)。心房颤动患者,miR-21与NT-proBNP呈正相关性,而miR-150与NT-proBNP呈负相关性(P<0.05)。心房颤动患者,miR-21、NT-proBNP水平在射频消融治疗术后显著降低。miR-150水平术后显著升高(P<0.05)。miR-21预测心房颤动射频消融术后复发的AUC值为0.737(0.625~0.850),miR-150预测心房颤动射频消融术后复发的AUC值为0.696(0.550~0.842)。结论miR-21高表达、miR-150低表达与心房颤动和NT-proBNP水平升高相关。miR-21水平在射频消融术后显著降低、而miR-150水平则术后显著升高。miR-21、miR-150表达水平对预测心房颤动射频消融术后的复发具有一定价值。 Objective To investigate the correlations of serum microRNA(miR-21,miR-150)levels with atrial fibrillation and NT-proBNP levels,as well as the changes after radiofrequency ablation of atrial fibrillation.Methods 103 patients with atrial fibrillation were enrolled,and 50 volunteers were enrolled in the same period as the control group.The expressions of miR-21 and miR-150 in the serum of the two groups were detected by real-time fluorescence quantitative PCR technology.At the same time,the plasma levels of N-terminal pro-brain natriuretic peptide(NT-proBNP)in the two groups of patients were also assessed.The correlations of miR-21 and miR-150 with atrial fibrillation and NT-proBNP levels were determined.At the same time,the expression changes of miR-21 and miR-150 and the level of NT-proBNP in patients with atrial fibrillation after radiofrequency ablation were analyzed.Results The levels of miR-21 and NT-proBNP in the atrial fibrillation group were significantly higher than those in the control group,and the level of miR-150 in the atrial fibrillation group was significantly lower than that in the control group(P<0.05).The levels of miR-21 and NT-proBNP in patients with persistent atrial fibrillation were significantly higher than those in patients with paroxysmal atrial fibrillation,and the level of miR-150 in patients with persistent atrial fibrillation was significantly lower than that in patients with paroxysmal atrial fibrillation(P<0.05).In patients with atrial fibrillation,miR-21 was positively correlated with NT-proBNP,and miR-150 was negatively correlated with NT-proBNP(P<0.05).In patients with atrial fibrillation,the levels of miR-21 and NT-proBNP were significantly decreased after radiofrequency ablation.The level of miR-150 was significantly increased after surgery(P<0.05).The AUC value of miR-21 for predicting the recurrence of atrial fibrillation after radiofrequency ablation was 0.737(0.625-0.850),and the AUC value of miR-150 for predicting the recurrence of atrial fibrillation after radiofrequency ablation was 0.696(0.550-0.842).Conclusion High expression of miR-21 and low expression of miR-150 are associated with atrial fibrillation and elevated NT-proBNP levels.The level of miR-21 is significantly reduced after radiofrequency ablation,while the level of miR-150 is significantly increased.The expression levels of miR-21 and miR-150 can predict the recurrence of atrial fibrillation after radiofrequency ablation.
