miR-1185-5p通过靶向PITPNB抑制鼻咽癌细胞的增殖和迁移

摘要 目的 探讨miR-1185-5p通过靶向磷脂酰肌醇转运蛋白β(PITPNB)对鼻咽癌CNE-2细胞增殖和迁移的影响。方法 应用TCGA数据库分析miR-1185-5p和PITPNB在头颈部鳞状细胞癌中的表达及PITPNB表达与病人总生存率的关系,CCK-8实验和划痕实验检测miR-1185-5p以及PITPNB对CNE-2细胞增殖和迁移的影响,TargetScan和双荧光素酶报告基因检测预测及验证miR-1185-5p和PITPNB之间的结合关系。结果 与癌旁正常组织相比,头颈鳞癌组织中miR-1185-5p表达下调(t=-2.17,P<0.05),PITPNB表达上调(Z=70,P<0.001);PITPNB高表达预示着病人较低的总生存率(χ^(2)=6.05,P<0.05);miR-1185-5p可抑制CNE-2细胞的增殖(F=8.086,P<0.05)和迁移(t=12.83,P<0.05),而PITPNB可促进CNE-2细胞的增殖(F=18.010,P<0.01)和迁移(t=13.39,P<0.05)。双荧光素酶报告基因检测验证了miR-1185-5p与PITPNB间的靶向结合关系(t=8.05,P<0.01)。结论 miR-1185-5p通过靶向PITPNB抑制鼻咽癌CNE-2细胞的增殖和迁移。 Objective To investigate the effect of miR-1185-5p on the proliferation and migration of nasopharyngeal carcinoma CNE-2 cells by targeting phosphatidylinositol transfer protein beta(PITPNB). Methods TCGA database was used to analyze the expression of miR-1185-5p and PITPNB in head and neck squamous cell carcinoma and the association between PITPNB and overall survival rate. CCK-8 assay and scratch assay were used to observe the effect of miR-1185-5p and PITPNB on the proli-feration and migration of CNE-2 cells, and TargetScan and dual-luciferase reporter assay were used to predict and validate the bin-ding relationship between miR-1185-5p and PITPNB. Results Compared with normal adjacent tissue, head and neck squamous cell carcinoma tissue had downregulated expression of miR-1185-5p(t=-2.17,P<0.05) and upregulated expression of PITPNB(Z=70,P<0.001). High expression of PITPNB predicted a relatively low overall survival rate of patients(χ^(2)=6.05,P<0.05). The results showed that miR-1185-5p inhibited the proliferation(F=8.086,P<0.05) and migration(t=12.83,P<0.05) of CNE-2 cells, while PITPNB promoted the proliferation(F=18.010,P<0.01) and migration(t=13.39,P<0.05) of CNE-2 cells. Dual-luciferase reporter assay validated the binding relationship between miR-1185-5p and PITPNB(t=8.05,P<0.01). ConclusionmiR-1185-5p inhibits the proliferation and migration of nasopharyngeal carcinoma CNE-2 cells by targeting PITPNB.
