七味葡萄散对哮喘模型大鼠血浆促肾上腺皮质激素水平的影响

摘要 目的研究七味葡萄散(QWPTS)对哮喘模型大鼠血浆促肾上腺皮质激素(ACTH)水平的影响。方法用卵清蛋白致敏Wistar大鼠的方法建立哮喘模型,按体重均一原则随机分为5组,每组10只,分别为空白对照组、哮喘模型组、QWPTS低、中、高剂量组。光镜下观察肺组织的病理学变化,用ELISA法检测大鼠血中ACTH的含量并进行比较。结果哮喘模型组大鼠肺泡壁结构受损、增厚,支气管腔狭窄,黏膜上皮细胞脱落,支气管壁和肺泡内可见大量炎性细胞浸润;QWPTS低剂量组肺组织未见明显的病理损伤,QWPTS中剂量组肺组织的损伤较哮喘模型组轻,QWPTS高剂量组较哮喘模型组重。哮喘模型组血浆ACTH水平低于空白对照组,差异有统计学意义(P<0.01);QWPTS低、中、高剂量组的血浆ACTH水平高于哮喘模型组,差异有统计学意义(P<0.05);QWPTS低、中、高剂量组的血浆ACTH水平两两比较,差异无统计学意义(P>0.05)。结论QWPTS可减轻哮喘模型大鼠的气道损伤,提高哮喘模型大鼠血浆ACTH水平,其机制可能与改善下丘脑-垂体-肾上腺皮质(HPA)轴功能有关。 Objective To study the effect of Qiwei Putaosan(QWPTS)on plasma adrenocorticotropic hormone(ACTH)level in asthma model rats.Methods Wistar rats were sensitized with obumin to establish asthma model,they were randomly divided into 5 groups according to the principle of equal body weight,with 10 rats in each group,including blank control group,asthma model control group,QWPTS low,medium and high dose groups.The pathological changes of lung tissue were observed under light microscope,and the content of ACTH in blood of rats was detected by ELISA.Results In the asthma model group,the structure of alveolar wall was damaged and thickened,the bronchial lumen was narrow,the mucosal epithelial cells were shed,and a large number of inflammatory cells were infiltrated in the bronchial wall and alveoli.No obvious pathological injury was observed in the lung tissue of the low-dose QWPTS group,the lung tissue injury of the medium-dose QWPTS group was lighter than that of the asthma model group,and the lung tissue injury of the high-dose QWPTS group was heavier than that of the asthma model group.The plasma ACTH level of asthma model group was lower than that of blank control group,and the difference was statistically significant(P<0.01).The plasma ACTH level in low,medium and high dose QWPTS groups was higher than that in asthma model group,and the difference was statistically significant(P<0.05).Pairwise comparison of plasma ACTH level in low,medium and high dose QWPTS groups showed no significant difference(P>0.05).Conclusion QWPTS can reduce airway injury and increase plasma ACTH level in asthma model rats,and the mechanism may be related to improving hypothalamic-pituitary-adrenocortical(HPA)axis function.
