摘要
目的探讨天麻4个酚性成分的组合物对LPS刺激致BV2细胞炎症模型的影响。方法复制LPS刺激致BV-2细胞炎症模型;以NO、TNF-α、IL-1β、IL-6、PGE2和IL-10的含量及BV-2细胞活化后膜蛋白CD16/32、CD206表达的变化为指标,考察天麻酚性成分组合物的作用。结果采用1μg/mL浓度的LPS刺激BV-2细胞24 h可成功复制BV2细胞炎症模型;阳性药物米诺环素(1μmol/L)、天麻酚性成分组合物(10μg/mL)能明显减少LPS刺激BV-2细胞释放NO、TNF-α、IL-6、PGE2的含量(P<0.05或P<0.01);与模型组比较,10 ng/mL的IL-4、1μmol/L的米诺环素和1、0.1、0.01μmol/L的天麻酚性成分组合物能下调膜蛋白CD16/32的表达(P<0.01),并上调膜蛋白CD206的表达(P<0.01),使CD206/CD16/32比值增大。结论天麻酚性成分组合物可明显抑制BV-2细胞炎症反应,并能促进促炎表型的M1型小胶质细胞转化为抗炎表型的M2型小胶质细胞。
Objective:To study the effects of the composition of four phenolic components of Gastrodia elata on the inflammation model of BV2 cells induced by LPS.Methods:The inflammation model of BV-2 cells induced by LPS stimulation was replicated.The contents of NO,TNF-α,IL-1β,IL-6,PGE2 and IL-10 and the expression of membrane proteins CD16/32 and CD206 after activation of BV-2 cells were used as indexes to study the effects of gastroelata phenol-based composition.Results:The inflammatory model of BV2 cells was successfully induced by LPS of 1μg/mL for 24 h.The positive drug minocycline(1μmol/L)and gastrotenol phenol-based compound(10μg/mL)could significantly reduce the contents of NO,TNF-α,IL-6 and PGE2 released by LPS-stimulated BV-2 cells(P<0.05 or P<0.01).Compared with that of the model group,10 ng/mL IL-4,1μmol/L minocycline and 1,0.1,0.01μmol/L gastrophenol-based composition down-regulated the expression of membrane protein CD16/32(P<0.01),and up-regulated the expression of membrane protein CD206(P<0.01).The ratio of CD206/CD16/32 was increased.Conclusion:Gastrodia phenol-based composition can significantly inhibit the inflammatory response of BV-2 cells and promote the transformation of M1-type microglia with pro-inflammatory phenotype into M2-type microglia with anti-inflammatory phenotype.
