摘要
目的建立苦皮藤高效液相色谱(HPLC)指纹图谱,并对其中儿茶素、表儿茶素和3,4,5-trimethoxyphyeny-β-D-glucopyranoside进行含量测定,为苦皮藤的质量控制提供参考。方法运用HPLC法构建苦皮藤药材的指纹图谱,并进行相似度评价,确认共有峰,与对照品比对指认共有峰,结合化学计量学方法对不同产地苦皮藤质量及其控制方法进行分析。采用HPLC法同时测定苦皮藤药材中儿茶素、表儿茶素和3,4,5-trimethoxyphyeny-β-D-glucopyranoside的含量。结果建立了12批苦皮藤的指纹图谱,标定了22个共有峰,并指认出3个共有峰,12批药材的相似度在0.856~0.969;聚类分析和主成分分析将12批药材聚为3类,主成分分析得到3个主成分的累计方差贡献率为91.009%;正交偏最小二乘法-判别分析筛选得到10个差异标志物,其中2个VIP较大的差异成分为儿茶素和表儿茶素。儿茶素、表儿茶素和3,4,5-trimethoxyphyenyl-β-D-glucopyranoside的平均回收率分别为97.97%、98.18%、95.72%,含量分别为1.076~2.289、0.835~2.105、0.337~0.765 mg·g^(-1)。结论建立了苦皮藤的HPLC指纹图谱及3个有效成分含量的测定方法,可为初步评价苦皮藤药材的质量提供依据。
Objective To establish an HPLC fingerprint of Celastrus angulatus Maxim.,and to determine catechin,epicatechin and 3,4,5-trimethoxyphyeny-β-D-glucopyranoside for the quality control of Celastrus angulatus Maxim..Methods HPLC fingerprint of Celastrus angulatus Maxim.was established for similarity evaluation on the basis of common peaks through chemical pattern recognition.Catechin,epicatechin and 3,4,5-trimethoxyphyeny-β-D-glucopyranoside were simultaneously determined.Results In the HPLC fingerprint,22 common peaks were marked,and 3 peaks were accurately identified.The similarity of the 12 batches of sample ranged from 0.856 to 0.969,which were clustered into 3 groups by cluster analysis and principal component analysis.The cumulative variance contribution rate of the three principal components was 91.009%.Ten differential markers were screened by orthogonal partial least squares-discriminant analysis.Among them,two VIP differential components were catechin and epicatechin.The average recoveries of catechin,epicatechin and 3,4,5-trimethoxyphyeny-β-D-glucopyranoside were 97.97%,98.18%and 95.72%;the content of them was 1.076~2.289,0.835~2.105,and 0.337~0.765 mg·g^(-1),respectively.Conclusion The HPLC fingerprint and the determination of 3 effective components are established,providing a scientific basis for the quality evaluation of Celastrus angulatus Maxim..
