摘要
目的观察miR-629在乳腺癌组织中的表达及其对乳腺癌细胞增殖的影响。方法选取2020年3月至2020年4月在蚌埠医学院第一附属医院手术切除的乳腺癌组织及其癌旁组织各40例,观察miR-629对乳腺癌细胞MCF-7增殖能力的影响,应用实时荧光定量PCR法检测miR-629的表达,采用CCK-8法检测MCF-7细胞增殖能力的变化,Western Blot检测MCF-7细胞PTEN和PI3K/AKT信号蛋白的表达。结果乳腺癌组织中miR-629的mRNA相对表达水平(1.70±0.15),与癌旁组织(0.40±0.04)比较,显著增加(t=52.182,P<0.001);miR-629敲低后对乳腺癌细胞MCF-7增殖的影响,与NC组(转染NC)细胞对比,IN组(转染miR-629-inhibitor)细胞在48 h和72 h的增殖速度下降,差异有统计学意义(P<0.05)。NC组细胞克隆形成数(101.67±11.93)明显高于IN组(64.00±11.53),差异有统计学意义(t=3.932,P<0.05)。Western Blot结果表明IN组细胞中PTEN的表达水平(1.31±0.11)较NC组细胞(0.44±0.03)明显升高(t=13.216,P<0.001)。而P-PI3K和P-AKT蛋白明显降低(P<0.05)。结论miR-629在乳腺癌组织中高表达,可能通过靶向PTEN介导PI3K/AKT通路,调控对MCF-7细胞增殖的影响。提示miR-629靶向调控PTEN的表达促进乳腺癌细胞的增殖,有望为乳腺癌的临床诊疗提供新的思路。
Objective To observe the expression of miR-629 in breast cancer tissues and its effect on the proliferation of breast cancer cells.Methods This study selected forty cases of breast cancer tissues and 40 cases of adjacent tissues resected from patients in The First Affiliated Hospital of Bengbu Medical College from March 2020 to April 2020.This study moved on to observe the effect of miR-629 on the proliferation of MCF-7 breast cancer cells.The expression of miR-629 was detected by real-time PCR.The change of proliferation ability of MCF-7 cells was detected by the method of CCK-8.And Western blot was employed to detect the protein expression of PTEN and PI3K/AKT signaling in MCF-7 cells.Results The relative mRNA expression(1.70±0.15)of miR-629 in breast cancer tissues was significantly higher than that(0.40±0.04)in adjacent tissues(t=52.182,P<0.001).In terms of the effect of miR-629 knockdown on proliferation of MCF-7 breast cancer cells,the IN group(transfected miR-629-inhibitor)had a decrease in the proliferation rate of cells at 48 h and 72 h in comparison with the NC group(transfected NC),indicating significant difference(P<0.05).The NC group had significantly more cells(101.67±11.93)formed by clone than the IN group(64.00±11.53),indicating statistically significant difference(t=3.932,P<0.05).The results of Western blot showed that the expression level of PTEN in the IN group(1.31±0.11)was significantly higher than that(0.44±0.03)in the NC group(t=13.216,P<0.001).The P-PI3K and P-AKT proteins significantly decreased(P<0.05).Conclusion miR-629 is highly expressed in breast cancer tissues,which may regulate the proliferation of MCF-7 cells by targeting PTEN to mediate PI3K/AKT pathway.These results suggest that miR-629 can promote the proliferation of breast cancer cells by target-regulating PTEN expression,which is expected to provide new ideas for the clinical diagnosis and treatment of breast cancer.
