转录组测序分析急、慢性乙型肝炎患者外周血单个核细胞基因变化

Transcriptome sequencing analysis of gene changes in peripheral blood mononuclear cells of patients with acute hepatitis B and chronic hepatitis B

摘要 目的筛选急性乙型肝炎(acutehepatitisB,AHB)和慢性乙型肝炎(chronichepatitisB,CHB)外周血单个核细胞中异常表达基因。方法收集在右江民族医学院附属医院就诊的3例AHB与3例CHB外周血,分离单个核细胞,提取RNA后进行转录组测序,根据P<0.05且|log2FC|>1计算两组样本的表达差异,绘制差异基因热图。对差异基因进行GO功能与KEGG信号通路聚类。结果AHB组和CHB组两组间满足P<0.05且|log2FC|>1差异条件的基因共计725个,其中上调372个,下调353个。差异基因主要富集的GO功能:微管发生功能、内源性刺激反应功能、组织发生功能、细胞增殖调控功能与细胞增殖功能和细胞外区域功能。差异基因主要富集的KEGG信号通路:PI3K-Akt信号通路、MAPK信号通路、硫胺素代谢通路、癌症转录失调和IL-17信号通路。结论提供了AHB和CHB外周血单个核细胞中差异表达基因变化的一般情况与可能调控功能和通路,可能有助于阐明AHB和CHB的发展机制。 Objective To screen differentially expressed genes in peripheral blood mononuclear cells of patients with acute hepatitis B(AHB)and those with chronic hepatitis B(CHB).Methods This study collected the peripheral blood from 3 patients with AHB and 3 patients with CHB in the Affiliated Hospital of Youjiang Medical University for Nationalities.The mononuclear cells in their blood were isolated for RNA extraction to perform transcriptome sequencing.The expression difference between two groups of samples was calculated according to P<0.05 and|log2FC|>1 and the heat map of differential genes was drawn.GO function and KEGG signaling pathway were clustered for differentially-expressed genes.Results In AHB group and CHB group,a total of 725 genes met the difference condition of P<0.05 and|log2FC|>1.Among these genes,372 were up-regulated and 353 genes down-regulated.The GO functions enriched in differentially-expressed genes included micro-tubule genesis,endogenous stimulus response,histogenesis,cell proliferation regulation,cell proliferation and extracellular domain.KEGG signaling pathways enriched in differentially expressed genes included PI3K-Akt signaling pathway,MAPK signaling pathway,thiamine metabolism pathway,cancer transcriptional dysregulation and IL-17 signaling pathway.Conclusion This study provides general information and possible regulatory functions and pathways of differentially-expressed genes in peripheral blood mononuclear cells of AHB and CHB.It may help elucidate the development mechanism of AHB and CHB.
