摘要
目的探讨内质网核糖体结合蛋白1(RRBP1)在肝癌组织细胞中的表达及在肝癌发展中的调控作用。方法分别用生物信息学分析与免疫组化实验探讨肝癌患者肝癌组织与癌旁正常组织中RRBP1的表达。用RT-PCR与Western blot实验检测体外肝癌细胞与正常肝细胞中RRBP1的表达。用小干扰RNA(siRNA)下调RRBP1表达后,分别用MTS实验、EdU实验与克隆形成实验检测肝癌细胞活力、增殖与克隆形成。结果肝癌组织中RRBP1表达显著高于癌旁正常组织(P<0.05)。RRBP1在Hep3B、HLE、HLF、SNU-739与SNU-368中的表达高于正常肝细胞LO2(P<0.05)。下调RRBP1后,肝癌细胞的活力、增殖与克隆形成能力均被显著抑制(P<0.05)。结论RRBP1在肝癌中异常高表达,RRBP1通过上调肝癌细胞的活力、增殖与克隆形成能力而促进肝癌的发展。
Objective To investigate the expression and biological roles of endoplasmic reticulum ribosome-binding protein 1(RRBP1)in the regulation of liver cancer cell growth.Methods Bioinformatics analysis and immunohistochemical staining were used to determine the expression of RRBP1 in tumor tissues and tumor-adjacent normal tissues of patients with liver cancer.Real-time polymerase chain reaction(RT-PCR)and western blot were performed to measure the expression of RRBP1 in human liver cancer cell lines and normal liver cell line.MTS assay,EdU assay and colony formation assay were used to test the viability,proliferation and colony formation of liver cancer cells after RRBP1 was knocked-down by small interference RNA(siRNA).Results The expression level of RRBP1 was significantly higher in tumor tissues than in tumor-adjacent normal tissues in patients with liver cancer(P<0.05).RRBP1 expression level was significantly higher in five liver cancer cell lines(Hep3B,HLE,HLF,SNU-739 and SNU-368)than in a normal liver cell line(LO2)(P<0.05).The viability,proliferation and colony formation abilities were markedly inhibited after RRBP1 was knocked-down in liver cancer cells(P<0.05).Conclusion RRBP1 is overexpressed in liver cancer,it promotes the development of liver cancer by enhancing cell viability,proliferation and colony formation abilities.
