豌豆多糖对DSS诱导结肠炎小鼠的保护作用

摘要 结肠炎发病率和流行率较高且其治疗药物存在一定副作用,因此人们逐渐寻求包括多糖在内的天然来源化合物作为治疗结肠炎的新手段。本研究采用水提醇沉提取、制备豌豆多糖,分析其基本理化性质特征,进一步构建葡聚糖硫酸钠(DSS)诱导小鼠溃疡性结肠炎模型,研究低、中、高(200,400,600 mg·kg-1小鼠体重)剂量豌豆多糖对小鼠溃疡性结肠炎的干预效果。结果显示,纯化后的豌豆多糖总糖含量可达85.1%,主要组分重均分子量为610 kDa,主要由Ara(20.1%)、Gal(15.1%)和GalA(13.1%)等单糖组成,是一种果胶类多糖。动物实验结果显示,相比于低剂量和高剂量,中剂量豌豆多糖表现出更好的保护作用,能够在一定程度上延缓体重下降和DAI值上升,抑制结肠缩短,降低结肠组织炎症浸润,保护其结构完整。豌豆多糖干预降低了促炎细胞因子IL-6、IL-1β水平,提高了抗炎细胞因子IL-10、IL-22(P<0.05)水平。综上所述,豌豆来源多糖是一种果胶类多糖,可通过调节炎症因子表达进而发挥对DSS诱导结肠炎小鼠的保护作用,上述结果为开发天然来源的结肠炎辅助治疗产品提供理论依据。 The incidence and prevalence of ulcerative colitis have a high incidence rate and the therapeutic drugs have certain side effects.Therefore, people are gradually seeking naturally derived compounds, including polysaccharides, as new treatments for colitis.In this study, the pea polysaccharides were extracted and prepared by water-extracted alcohol sinks, and their basic physicochemical properties were analyzed.Then, a model of dextran sodium sulfate(DSS) induced ulcerative colitis in mice was constructed, and the intervention effects of low, medium and high(200,400,600 mg·kg-1mouse body weight) doses of pea polysaccharides on ulcerative colitis in mice were studied.The results showed that the total sugar content of purified pea polysaccharides could reach 85.1%,and the average molecular weight of the main components was 610 kDa.Furthermore, it was a pectin polysaccharide, mainly composed of monosaccharide such as Ara(20.1%),Gal(15.1%) and GalA(13.1%).The results of animal experiments showed that compared with low doses and high doses, medium doses of pea polysaccharides showed better protective effect.Also, they could delay the loss of weight and the rise of DAI,inhibit the shortening of colon, reduce the inflammatory infiltration of colon tissue, and protect its structural integrity.The pea polysaccharides intervention reduced the levels of pro-inflammatory cytokines IL-6 and IL-1β,and increased the levels of anti-inflammatory cytokines IL-10 and IL-22(P<0.05).In summary, pea-derived polysaccharides are pectin polysaccharides that can play a protective role in mice with DSS-induced colitis by regulating the expression of inflammatory factors.These results provide a theoretical basis for the development of adjuvant treatment products of natural origin for colitis.
