Beclin1在大鼠糖尿病溃疡创面组织中表达变化

摘要 目的观察自噬相关蛋白(Beclin)1在糖尿病大鼠溃疡创面愈合中的变化,初步探讨Beclin1与糖尿病慢性创面愈合的关系。方法SPF级Wistar雄性大鼠18只,随机分为对照组、模型组各9只。对照组建立常规创面模型,模型组建立糖尿病创面模型。两组分别于模型建立后第1天、第11天、第18天观察创面愈合情况、组织苏木素-伊红(HE)染色病理改变、Beclin1蛋白及其磷酸化水平、Beclin1基因表达。结果对照组创面愈合效率、组织病理改善均优于模型组(P<0.05);对照组Beclin1、p-Beclin1蛋白及Beclin1 mRNA在创面愈合早期的表达高于模型组,而在愈合晚期的表达低于模型组,差异均有统计学意义(P<0.05)。结论Beclin1在糖尿病慢性创面组织中低表达,可能是糖尿病溃疡创面愈合不良的机制之一。 Objective To observe the changes of autophagy associated protein(Beclin)1 in the process of wound healing in diabetic rats,and to preliminarily study on the relationship between Beclin1 and chronic wound healing in diabetes mellitus.Methods Eighteen male SPF-grade Wistar rats were randomly divided into control group and model group,with 9 rats in each group.Conventional wound model was established in the control group while diabetic wound model was established in the model group.The wound healing,histopathological changes under htoxylin eosin(HE)staining,Beclin1 protein and its phosphorylation level and Beclin1 gene expression were observed on day 1,11 and 18 in two groups after the model were established.Results The wound healing efficiency and histopathological improvement in the control group were better than those in the model group(P<0.05).The expressions of Beclin1,p-Beclin1 protein and Beclin1 mRNA in the control group in the early stage of wound healing were higher than those in the model group,while the expressions in the late stage of wound healing were lower than those in the model group,the differences were statistically significant(P<0.05).Conclusions The low expression of Beclin1 in diabetic chronic wound tissues may be one of the mechanisms of poor wound healing in diabetic ulcer.
