表面脱钙法在肺腺癌骨转移组织制片中的应用

摘要 目的探讨表面脱钙法在肺腺癌骨转移组织中的应用效果。方法选取2020年6月至2022年4月在福州市第二医院手术的20例肺腺癌骨转移标本作为研究对象,切取同一份标本得到相同体积组织3份,分为表面脱钙组、常规脱钙组、不脱钙组。比较表面脱钙组和常规脱钙组的脱钙时间,比较三组的HE染色与免疫组化染色质量。结果表面脱钙组的脱钙时间为(0.21±0.05)h,短于常规脱钙组的(2.45±0.76)h,差异有统计学意义(P<0.05)。表面脱钙组、常规脱钙组、不脱钙组的HE染色得分分别为(9.45±0.99)、(9.40±1.03)、(7.70±1.47)分,三组比较差异有统计学意义(P<0.05),且表面脱钙组、常规脱钙组的HE染色得分高于不脱钙组,差异有统计学意义(P<0.05)。三组的Ki-67、TTF-1、Napsin-A免疫组化染色评分比较,差异均有统计学意义(P<0.05),且表面脱钙组的Ki-67、TTF-1、Napsin-A免疫组化染色评分高于常规脱钙组、不脱钙组,差异有统计学意义(P<0.05)。结论肺腺癌骨转移标本采用表面脱钙法脱钙时间更短,HE染色和免疫组化染色质量更佳,值得推广使用。 Objective To study the effect of surface decalcification in bone metastasis of lung adenocarcinoma.Methods Twenty lung adenocarcinoma bone metastatic specimens operated in the Fuzhou Second Hospital from June 2020 to April 2022 were selected as the research object,and three tissues of the same volume were obtained from the same specimen,which were divided into surface decalcification group,routine decalcification group and non-decalcification group.The decalcification time of surface decalcification group and routine decalcification group was compared,and the quality of HE staining and immunohistochemical staining among the three groups was compared.Results The decalcification time in the surface decalcification group was(0.21±0.05)h,which was shorter than that in the conventional decalcification group(2.45±0.76)h,and the difference was statistically significant(P<0.05).The HE staining scores of the surface decalcification group,the routine decalcification group and the non-decalcification group were(9.45±0.99),(9.40±1.03)and(7.70±1.47),respectively,and the difference among the three groups was statistically significant(P<0.05),and the HE staining scores of the surface decalcification group and the routine decalcification group were higher than those of the non-decalcification group,the differences were statistically significant(P<0.05).The immunohistochemical staining scores of Ki-67,TTF-1 and Napsin-A of the three groups were statistically significant(P<0.05),and the immunohistochemical staining scores of Ki-67,TTF-1 and Napsin-A of the surface decalcification group were higher than those of the conventional decalcification group and the non-decalcification group,with statistically significant differences(P<0.05).Conclusion The surface decalcification method for bone metastases of lung adenocarcinoma has shorter decalcification time and better HE and immunohistochemical staining quality,which is worthy of popularization and application.
