淫羊藿苷配伍人参皂苷Rg1对骨髓间充质干细胞成骨分化的作用 被引量:1

Effects of Icariin Combined with Ginsenoside Rg1 on Osteogenic Differentiation of Bone Marrow Mesenchymal Stem Cells

摘要 研究淫羊藿苷和人参皂苷Rg1不同浓度配伍对骨髓间充质干细胞(BMMSCs)成骨分化的作用。采用Cell Counting Kit-8(CCK-8试剂盒)观察淫羊藿苷和人参皂苷Rg1单独用药及不同浓度配伍对C57小鼠骨髓间充质干细胞增殖情况的影响;在成骨诱导第16、30 d后用茜素红进行染色,观察成骨细胞矿化能力;采用反转录实时定量PCR(RT-PCR)及蛋白质印记(WB)检测成骨细胞标记性蛋白人骨形态发生蛋白BMP2、RUNX2、OPN、OCN的表达情况。淫羊藿苷和人参皂苷Rg1单独用药最佳浓度分别为1μmol/L和10μmol/L;两者配伍最佳浓度分别为1μmol/L和20μmol/L。与单独用药相比,配伍能够明显促进诱导细胞的钙化作用并且上调成骨细胞标记性蛋白BMP2、OCN、OPN、RUNX2等的表达,淫羊藿苷和人参皂苷Rg1配伍能够促进骨髓间充质干细胞成骨分化。 To explore the effect of different dose combination of icariin and ginsenoside Rg1 on osteogenic differentiation of BMMSCs.CCK-8 was applied to observe the effects of icariin and ginsenoside Rg1 single dose and different concentrations of compatibility on the proliferation of BMSCs under common culture conditions. Alizarin red staining method was used to observe calcification after 16 days and 30 days of osteogenic induction. RT-PCR and WB were used to detect the expression of osteoblast marker proteins BMP2,OCN,OPN and RUNX2. The optimal concentrations of icariin and ginsenoside Rg1 monomer are 1 μmol/L and 10 μmol/L respectively,While their combined concentrations are 1 μmol/L and 20 μmol/L as the best compatibility. Compared with the drug alone,the combination of icariin and ginsenoside Rg1 can significantly promote the induction of calcification and up-regulate the expression of BMP2,OCN,OPN and RUNX2. Combination of icariin and ginsenoside Rg1 can promote osteogenic differentiation of BMMSCs.
