摘要
背景:骨性关节炎是最常见的关节疾病之一,随着骨性关节炎的全球发病率和患病率不断上升,有效早期诊断和治疗骨性关节炎已成为一个紧迫的问题。目的:阐述长链非编码RNA、环状RNA和假基因的重要生物学功能以及它们作为内源性竞争RNA在骨性关节炎发病中的调控作用,以获得内源性竞争RNA参与骨性关节炎进展全面、深入和新的理解,为骨性关节炎的早期诊断和治疗提供新线索。方法:检索中国知网、PubMed、维普和万方数据库2022年前发表的文献,中文检索词为“骨性关节炎、内源性竞争RNA、长链非编码RNA、环状RNA、假基因”,英文检索词为“osteoarthritis,competing endogenous RNA,circular RNA,long non-coding RNAs,pseudogenes”。结果与结论:(1)具有相同微小RNA反应元件的非编码RNA,包括长链非编码RNA、环状RNA和假基因共同参与构建了以微小RNA为中心的内源性竞争RNA调控网络;(2)非编码RNA、环状RNA和假基因可作为内源性竞争RNA,通过“海绵”作用吸附微小RNA进而调控基因表达;(3)长链非编码RNA、环状RNA和假基因可作为内源性竞争RNA参与调控骨性关节炎软骨细胞的增殖、凋亡和自噬、细胞外基质的合成与降解以及炎症事件的发生;(4)内源性竞争RNA调控网络切实运用到临床上还面临诸多挑战和问题;(5)内源性竞争RNA作为骨性关节炎诊断标记物或治疗靶点的研究仍处于早期阶段,目前对内源性竞争RNA调节网络的研究主要还集中在单个内源性竞争RNA的鉴定和验证上,该网络有待进一步补充和完善;但随着先进技术的发展,内源性竞争RNA有望成为骨性关节炎疾病的早期诊断标记物和治疗靶点。
BACKGROUND:Osteoarthritis is one of the most common joint diseases.With the increasing global incidence and prevalence of osteoarthritis,effective early diagnosis and treatment of osteoarthritis have become an urgent problem.OBJECTIVE:To elu cidate the important biological functions of long non-coding RNAs,circular RNAs and pseudogenes and their regulatory roles as competing endogenous RNAs in osteoarthritis pathogenesis to gain a comprehensive,in-depth and new understanding of the involvement of competing endogenous RNAs in osteoarthritis progression and to provide new clues for early diagnosis and treatment of osteoarthritis.METHODS:CNKI,PubMed,VIP and WanFang databases we re searched for articles published before 2022.The search terms were"osteoarthritis,competing endogenous RNA,circular RNA,long non-coding RNAs,pseudogenes"in Chinese and English.RESULTS AND CONCLUSION:Non-coding RNAs with the same mic roRNA response elements,including long non-coding RNAs,cyclic RNAs and pseudogenes,are involved in the construction of a regulatory network of competing endogenous RNAs cente red on microRNAs.Non-coding RNAs,cyclic RNAs and pseudogenes can act as competing endogenous RNAs to adsorb microRNAs through"sponge"so as to regulate gene expression.Long non-coding RNAs,cyclic RNAs and pseudogenes can act as competing endogenous RNAs to regulate the proliferation,apoptosis and autophagy of osteoarthritic chondrocytes,the synthesis and degradation of extra cellular matrix and the occurrence of inflammato ry events.The practical application of the regulatory network of competing endogenous RNA to the clinical setting still fa ces many challenges and problems.The research on competing endogenous RNA as diagnostic marke rs or therapeutic targets for osteoarthritis is still at an early stage,and the current research on competing endogenous RNA regulato ry network is still focused on the identification and validation of individual competi ng endogenous RNAs,and this regulatory network needs to be further supplemented and improved.Howeve r,with the development of advanced technologies,competing endogenous RNAs are expected to become early diagnostic marke rs and therapeutic targets for osteoarthritis diseases.
