基于功能性便秘小鼠结肠AQP3表达探讨助阳通便膏治疗机制

Effect of Zhuyang Tongbian Ointment(助阳通便膏)on Aquaporin3of Mice with Functional Constipation

目的 通过予以慢性便秘模型小鼠助阳通便膏治疗,研究助阳通便膏对功能性便秘模型小鼠结肠水通道蛋白3(AQP3)的影响。同时探讨助阳通便膏对功能性便秘的作用机制。方法 将60只小鼠随机分为正常组、模型组、低剂量组、中剂量组及高剂量组,每组12只。除正常组小鼠外,其余小鼠均予以盐酸洛哌丁胺灌胃建立模型,并分应用相应药物干预,观察治疗作用。所有小鼠均采用免疫组化和RT-PCR的方法检测小鼠结肠AQP3的分布和表达情况。结果 与模型组比较,低、中、高剂量组首次排黑便时间明显缩短,24 h排便重量、粪便含水量均明显增加(P<0.05),AQP3含量及表达均减少(P<0.05);高剂量组较中剂量组AQP3含量及表达略降低,但无明显差异(P>0.05)。结论 助阳通便膏能有效改善便秘症状。助阳通便膏可以降低AQP3表达从而减少肠道水分吸收,可能是其治疗功能性便秘的作用机制之一,但高剂量应用效果未明显增加。

Objective To study the effect of Zhuyang Tongbian Ointment(助阳通便膏)on colonic aquaporin 3(AQP3)in mice with functional constipation.At the same time,to explore the mechanism of Zhuyang Tongbian Ointment on functional constipation.Methods Sixty mice were randomly divided into normal group,model group,low dose group,medium dose group and high dose group,with 12 mice in each group.Except the normal mice,the other mice were given loperamide hydrochloride by gavage to establish the model,and were intervened with the corresponding drugs to observe the therapeutic effect.The distribution and expression of AQP3 in the colon of mice were detected by immunohistochemistry and RT-PCR.Results Compared with those of the model group,the first time of black stool excretion in the low,medium and high dose groups was significantly shortened,the 24-hour defecation weight and fecal water content were significantly increased(P<0.05),and the content and expression of AQP3 were decreased(P<0.05).The content and expression of AQP3 in the high dose group were slightly lower than that in the medium dose group,but there was no significant difference(P>0.05).Conclusion Zhuyang Tongbian Ointment can effectively improve the symptoms of constipation.Zhuyang Tongbian Ointment can reduce the expression of AQP3 and reduce intestinal water absorption,which may be one of its mechanisms in the treatment of functional constipation,but the effect of high dose application did not increase significantly.
