基于肠促胰岛素激素水平变化探讨消渴健脾方对脾虚湿盛型2型糖尿病SD大鼠的降糖作用

Hypoglycemic Effect of Xiaoke Jianpi Prescription on Type 2 Diabetes Mellitus SD Rats with Spleen Deficiency and Dampness Exuberance:A Study Based on the Changes of Incretin Hormones

目的探究基于肠促胰岛素激素水平变化探讨消渴健脾方对脾虚湿盛型2型糖尿病SD大鼠的降糖机制。方法将SD大鼠随机分为空白对照组(Control组),粪便移植对照组(Fecal组),粪便移植+脾虚湿盛型2型糖尿病大鼠(DM组),粪便移植+脾虚湿盛型2型糖尿病大鼠A组+二甲双胍缓释片(DM+melbine组),粪便移植+脾虚湿盛型2型糖尿病大鼠B组+消渴健脾方(DM+QK组),比较各组治疗前后SD大鼠一般情况观察,ELISA检测胰高糖素样肽-1(Glucagon-like peptide-1,GLP-1)、葡萄糖依赖性促胰岛素多肽(Gastric inhibitory polypeptide,GIP)、胰岛素(Insulin,INS)、葡萄糖(Glucose,GLU)的表达变化及胰岛素抵抗指数(HOMA-IR)的变化。结果消渴健脾方可以有效改善脾虚湿盛型2型糖尿病SD大鼠的临床不良症状;从时间节点上发现DM+melbine组的药效具有一定的时间依赖性,可以有效地提高GLP-1和GIP的表达量,且与melbine组药效相近;DM+melbine组和DM+QK组可以有效降低模型的GLU与INS的表达。DM+QK组的GLU和INS表达在120 min时与Control组相似,效果要优于melbine;DM+melbine组和DM+QK组的可以有效降低模型的HOMA-IR值。结论消渴健脾方可降低T2DM SD大鼠的血糖、胰岛素、胰岛素抵抗,促进GIP、GLP-1水平恢复,同时药物的疗效稳定性与有效性较高。

Objective To investigate the hypoglycemic mechanism of Xiaoke Jianpi Prescription against type 2 diabetes mellitus SD rats with spleen deficiency and dampness exuberance based on the changes of incretin hormones.Methods SD rats were randomly divided into blank control group(control group),fecal transplantation control group(fecal group),fecal transplantation+type 2 diabetes mellitus rats with spleen deficiency and dampness exuberance(DM group),fecal transplantation+type 2 diabetes mellitus rats with spleen deficiency and dampness exuberance A group+metformin sustained-release tablets(DM+melbine group),and fecal transplantation+type 2 diabetes mellitus rats with spleen deficiency and dampness exuberance B group+Xiaoke Jianpi Prescription(DM+XJ group).The general situation of SD rats before and after treatment in each group was observed and compared.Enzyme-linked immunosorbent assay(ELISA)was used to detect the expressions of glucagon-like peptide-1(GLP-1),glucose-dependent insulinotropic polypeptide(GIP),insulin(INS),and glucose(GLU),and the changes in homeostatic model assessment of insulin resistance(HOMA-IR)value.Results Xiaoke Jianpi Prescription effectively alleviated the clinical adverse symptoms of type 2 diabetes mellitus SD rats with spleen deficiency and dampness exuberance.From the time points,it was found that the DM+melbine group had a certain time-dependent efficacy,with up-regulated expressions of GLP-1 and GIP,and XiaokeJianpi Prescription had similar efficacy to metformin sustained-release tablets.The DM+melbine group and DM+XJ group had reduced expressions of GLU and INS.The expressions of GLU and INS in the DM+XJ group were similar to those in the control group at 120 min,and the effect of Xiaoke Jianpi Prescription was better than that of metformin sustained-release tablets.The DM+melbine group and DM+XJ group had decreased HOMA-IR value and similar results.Conclusion Xiaoke Jianpi Prescription can effectively lower the GLU,INS,and INS resistance in type 2 diabetes mellitus SD rats and restore the GIP and GLP-1 levels,with high stability and efficacy.
