摘要 目的探讨美他多辛对大鼠酒精性肝病的预防作用。方法将30只SD大鼠随机均分为正常组、酒精性肝病模型组和美他多辛预防组,每组各10只。正常组喂养饲料,模型组喂饲料+52%白酒(8 g·kg^(-1)·d^(-1),2次/天)灌胃,美他多辛预防组喂养饲料+52%白酒(8 g·kg^(-1)·d^(-1),2次/天)灌胃+美他多辛溶液灌胃(300 mg·kg^(-1)·d^(-1),1次/天),至第4周末。第5周起,模型组及预防组两组白酒灌胃量提升(10 g·kg^(-1)·d^(-1))。第8周末,处死动物分离血清测肝功指标谷丙转氨酶(ALT)、谷草转氨酶(AST)、甘油三酯(TG)和总胆固醇(T-CHOL),氧化应激指标检测还原型谷胱甘肽(glutathione,GSH)、丙二醛(MDA)和超氧化物歧化酶(SOD)以及病理切片观察。结果酒精性肝病模型组血清ALT、AST、TG和T-CHOL升高,预防组ALT、AST水平均低于模型组(P<0.05)。预防组血清TG水平均低于模型组(P<0.05)。酒精性肝病模型组GSH、MDA和SOD升高,预防组血清SOD、血浆GSH水平均高于模型组(P<0.05),血清MDA水平低于模型组(P<0.05)。病理切片,预防组肝细胞脂肪变性及水肿有很大改善,肝索排列也较模型组组整齐,炎性细胞减少,未见大量坏死。结论美他多辛可以明显减轻血清中MDA、AST、CHOL、TG和ALT的水平,提升血浆GSH、血清SOD活性。 Objective To investigate the preventive effect of metadoxine on alcoholic liver disease in rats.Methods Thirty SD rats were randomly divided into normal group,alcoholic liver disease model group and metadoxine prevention group,with 10 rats in each group.The rats in normal group were fed with diet,the model group was fed with diet and 52%liquor(8 g·kg^(-1)·d^(-1),twice a day)by gavage,and the metadoxine prevention group was fed with diet and 52%liquor(8 g·kg^(-1)·d^(-1),twice a day)by gavage and metadoxine solution by gavage(300 mg·kg^(-1)·d^(-1),once a day)until the end of the 4th week.Since the 5th week,the amount of liquor gavage increased in the model group and the prevention group(10 g·kg^(-1)·d^(-1)).At the end of the 8th week,the animals were sacrificed to separate serum to measure liver function parameters including alanine aminotransferase(ALT),aspartate aminotransferase(AST),triglyceride(TG)and total cholesterol(T-CHOL),and oxidative stress parameters including reduced glutathione(GSH),malondialdehyde(MDA)and superoxide dismutase(SOD)and pathological section observation.Results Serum ALT,AST,TG and T-CHOL were increased in alcoholic liver disease model group,and ALT and AST levels in prevention group were significantly lower than those in model group(P<0.05).Serum TG levels in the prevention group were significantly lower than those in the model group(P<0.05).GSH,MDA and SOD were increased in the alcoholic liver disease model group,and the serum SOD and plasma GSH levels in the prevention group were significantly higher than those in the model group(P<0.05),and the serum MDA level was significantly lower than that in the model group(P<0.05).Pathological sections showed that hepatocyte steatosis and edema were greatly improved in the prevention group,hepatic cord arrangement was also neat compared with the model group,inflammatory cells were reduced,and no massive necrosis was observed.Conclusions Metadoxine could significantly reduces the levels of MDA,AST,CHOL,TG and ALT in serum,and increases the activities of GSH and SOD in plasma.
