摘要
目的 探讨丝聚蛋白(filaggrin, FLG)、分泌型卷曲相关蛋白1(secreted frizzled-related protein-1,SFRP1)、转化生长因子β1(transforming growth factor β1,TGF-β1)在口腔黏膜下纤维化(oral submucous fibrosis,OSF)患者中的表达。方法 选取OSF患者70例,依据病理学分为OSF早期组、中期组、晚期组,另选取同时段健康人群27例,作为正常组。对四组研究对象FLG、SFRP1、TGF-β1表达水平进行检测,使用免疫组化SP法检测FLG水平;使用酶联免疫吸附法检测SFRP1水平;使用地高辛标记TGF-β1 DNA探针检测TGF-β1表达水平。分析上述指标在OSF中的相关性。对分析中具有统计学意义的指标进行多指标Logistics回归分析。结果OSF早期组患者FLG水平高于正常组,OSF中期组患者FLG水平高于正常组、OSF早期组,OSF晚期组患者FLG水平高于其他三组(P<0.05)。OSF早期组患者SFRP1表达高于正常组,OSF中期组患者SFRP1表达高于正常组、OSF早期组,OSF晚期组患者SFRP1表达高于其他三组(P <0.05)。OSF早期组患者TGF-β1水平高于正常组,OSF中期组患者TGF-β1水平高于正常组、OSF早期组,OSF晚期组患者TGF-β1水平高于其他三组(P<0.05)。FLG、分泌型卷曲相关蛋白1、转化生长因子β1之间呈正相关。FLG、分泌型卷曲相关蛋白1、转化生长因子β1水平与OSF严重程度具有相关性。结论 FLG、SFRP1、TGF-β1在OSF患者中均呈异常高表达,并且具有一定的相关性。对FLG、分泌型卷曲相关蛋白1、转化生长因子β1指标进行检测,对OSF严重程度的临床诊断具有一定帮助。
Objective To investigate the expression of filaggrin(Flg), secretory frizzled related protein-1(SFRP1),transforming growth factor β1(TGF-β1) in patients with oral submucous fibrosis(OSF). Methods A total of 70 OSF patients were selected and classified into the early, middle, and late OSF groups based on the pathology, and 27 healthy individuals served as normal control group.Flg, SFRP1, and TGF-β1 expression levels were determined using IHC sp for flg, ELISA for SFRP1, and digoxigenin labeled TGF-β1 DNA probe for TGF-β1 expression.The correlation of the above indicators in OSF was analyzed. Results The level of FLG in OSF early group was higher than that in normal group, was higher in middle OSF group than that in normal group and early OSF group, was higher in late OSF group than that in other three groups(P<0.05).The expression of SFRP1 was higher in early OSF group than that in normal group, was higher in middle OSF group than that in normal group and early OSF group. The expression of SFRP1 in late OSF group was higher than that in other three groups(P<0.05). The level of TGF-β1 in early OSF group was higher than that in normal group, in middle OSF group was higher than that in normal group and early OSF group, was higher in late OSF group than that in other three groups(P<0.05).Flg, secretory curl associated protein 1 and transforming growth factor β1 were positively correlated. Conclusions Flg,SFRP1, TGF-β1 were abnormally expressed in OSF patients.The detection of FLG, secreted frizzled related protein 1, and transforming growth factor beta 1 indicators can provide some help in the clinical diagnosis of the severity of OSF.
