PRC1在肾细胞癌中的表达及临床意义研究

Expression and clinical significance of PRC1 in renal cell carcinoma

摘要 目的通过检测肾细胞癌(RCC)组织和配对癌旁组织中细胞质分裂调控蛋白1(PRC1)的表达,分析其表达水平与患者临床特征之间的关系,从而为PRC1的后续功能研究奠定基础。方法收集2010年1月至2016年12月深圳市第二人民医院的40例手术切除的RCC组织和癌旁组织标本,同时培养ACHN、786-O和HK-2细胞系,提取组织和细胞中的总RNA并逆转录合成cDNA,通过实时荧光定量PCR(qRT-PCR)技术检测PRC1的表达,统计分析其表达水平与患者临床病理特征之间的关系。结果ACHN、786-O细胞中的PRC1表达均高于HK-2细胞(均P<0.05)。RCC组织中PRC1的表达较癌旁组织明显升高(P<0.05)。PRC1的表达与患者的性别、年龄、肿瘤临床病理(TNM)分期、美国癌症联合会(AJCC)分期及Fuhrman分级均无相关性(均P>0.05)。结论PRC1在RCC组织及细胞中的表达水平升高,提示其与RCC发生机制有一定关系。 Objective To explore the expression of protein regulator of cytokinesis(PRC1)in renal cell carcinoma(RCC)and paired adjacent normal renal tissues,and analysize the relationships between the PRC1 expression and clinical features.Methods Total RNA extracted from 40 paired tissues obtained from nephrectomy were used to get cDNA by performing reverse transcription from January 2010 to December 2016 in our hospital.and ACHN,786-O,HK-2 cell lines were cultuzed.The expression of PRC1 was determined by quantitative real-time PCR(qRT-PCR)and the relationships with the clinical characteristics were explored by statistical analysis.Results The expression of PRC1 in ACHN and 786-O cell lines was higher than that in HK-2 cell lines(all P<0.05).The expression of PRC1 in RCC tissues was significantly higher than that in paracancerous tissues(P<0.05).The expression of PRC1 was not correlated with gender,age,TNM stage,American Joint Committee on Cancer(AJCC)stage,and Fuhrman grade(all P>0.05).Conclusions PRC1 expression was increased in RCC tissues and cell lines,indicating that it is potentially involved in the tumorigenesis of RCC.
