糙叶五加根皮和茎脂溶性成分的GC-MS分析及其抗炎活性

Analysis of the fat-soluble components from root bark and stem of Acanthopanax henryi(Oliv.) Harms by GC-MS and their anti-inflammatory activities

摘要 目的:采用气相色谱-质谱(Gas chromatography-mass spectrometry,GC-MS)联用法分析研究糙叶五加根皮和茎的脂溶性化学成分,并评价其抗炎活性。方法:采用乙醇加热回流提取、石油醚萃取的方法分别提取糙叶五加根皮和茎中的脂溶性化合物,提取液经挥干后使用二氯甲烷溶解,采用气相色谱-质谱联用技术结合质谱库检索系统对其脂溶性化学成分进行分离和鉴定,应用色谱峰面积归一化法计算各组分的相对百分含量;采用MTT法测试细胞毒性,Griess法检测炎症因子一氧化氮(NO)的抑制效果。结果:从糙叶五加根皮和茎的脂溶性成分中共鉴定出66个化合物,其中根皮和茎中分别鉴定出54种和15种脂溶性成分,分别占总峰面积的89.32%和82.33%。另外,在测试浓度为50μg·mL^(-1)时,糙叶五加根皮和茎的脂溶性成分对RAW264.7和BV2细胞均无细胞毒性,且对NO的产生具有较好的抑制活性。结论:糙叶五加根皮和茎的脂溶性成分均具有潜在的抗炎活性,然而,其化学成分种类和含量存在较大差异,使用时应加以区分,本研究为糙叶五加的质量评价和进一步的资源开发利用提供实验依据和理论参考。 Objective:To analyze the fat-soluble components from root bark and stem of A.henryi by gas chromatographymass spectrometry (GC-MS) and evaluate their anti-inflammatory activities.Methods:The fat-soluble components of root bark and stem of A.henryi were extracted by ethanol heating reflux extraction and petroleum ether extraction methods.The extract was volatilized and dissolved with dichloromethane.The fat-soluble chemical components were separated and identified by GC-MS and mass spectrometry library retrieval system.The relative percentage content of each compound was calculated by area normalization method.MTT and Griess assays were used to detect cytotoxicity and the inflammatory factor,nitric oxide (NO) inhibitory effect,respectively.Results:Sixty-six compounds were identified from the fat-soluble components of the root bark and stem of A.henryi,of which,54 and 15 fat-soluble chemical composition were identified in the root bark and stem,accounting for 89.32%and 82.33%of the total peak area,respectively.Furthermore,the fat-soluble components of the root bark and stem of A.henryi showed moderate NO inhibitory effects on RAW264.7 and BV2 cells at the concentration of 50μg·mL^(-1),with no cytotoxicity.Conclusion:The liposoluble components in the root bark and stem of A.henryi have potential anti-inflammatory activities.However,there are great differences in the types and contents of their chemical components,which should be distinguished when used.This study provides experimental basis and theoretical reference for the quality evaluation and further resource development and utilization of A.henryi.
