多囊卵巢综合征患者颗粒细胞蛋白质组学分析

摘要 目的应用定量蛋白质组学方法分析多囊卵巢综合征(PCOS)患者颗粒细胞蛋白质表达变化。方法收集2020年9月至2021年9月于我院生殖医学中心行IVF/ICSI助孕的患者取卵后废弃卵泡液标本24例,其中PCOS组12例和正常排卵对照组12例,从中提取颗粒细胞。采用液相色谱-串联质谱方法得到其蛋白质表达谱,生物信息学分析鉴定差异表达蛋白及其功能。采用Western Blot法验证质谱结果。结果共鉴定出329种差异表达蛋白,其中上调蛋白169个,下调蛋白160个。KEGG富集分析结果显示,差异蛋白在亮氨酸、异亮氨酸和缬氨酸分解,糖酵解/糖异生,谷胱甘肽代谢等通路显著富集。结论PCOS女性与正常排卵女性之间颗粒细胞蛋白质表达谱存在显著差异,进一步探索差异蛋白的功能或许可以为PCOS发病机制的研究提供线索。 Objective:To analyze the protein expression of granulosa cells in patients with polycystic ovary syndrome(PCOS)by quantitative proteomics.Methods:Twenty-four discarded follicular fluid samples were collected from the patients who underwent IVF/ICSI treatment in the Reproductive Medicine Center of the First Affiliated Hospital of Zhengzhou University from September 2020 to September 2021,including 12 samples in PCOS group and 12 samples in normal ovulation control group,and the granulosa cells were isolated from them.The protein expression profiles of granulosa cells were obtained by tandem mass spectrometry,and bioinformatics analysis was performed to identify differentially expressed proteins and their biological functions.Western blot was conducted to confirm the results of mass spectrometry.Results:A total of 329 differentially expressed proteins were identified,including 169 up-regulated proteins and 160 down-regulated proteins.The results of KEGG enrichment analysis showed that the differential proteins were significantly enriched in leucine,isoleucine and valine decomposition,glycolysis/gluconeogenesis,glutathione metabolism and other pathways.Conclusions:There are significant differences in the protein expression profiles of granulosa cells between PCOS women and normal ovulation women.Further exploration of the function of the differential proteins may provide clues for the study of the pathogenesis of PCOS.
