摘要 选用一组高效SSR引物,用于澳洲坚果杂交优株及诱变优株的DNA分子标记数据采集。从早期设计并筛选的SSR标记中挑选多态性高、重复性好、在染色体上分布均匀的9对引物,经PCR扩增,利用毛细管电泳,对1份化学诱变优株、52份杂交优株及13个亲本共66份材料进行电泳检测。1对引物在66份材料中存在较高的数据缺失率,最终选取7对SSR引物用于后续数据的采集。将7个亲本选为参照品种,并设置了7个亲本的两组平行试验,建立了53份优良单株及其亲本的分子标记数据库,可为其新品种登记提供技术支持。 A group of SSR markers were selected to collect DNA molecular marker data of Macadamia hybrid and mutated superior plants,for protecting breeding rights and interests.From the SSR markers screened in the early stage,9 pairs of primers with high polymorphism,good repeatability and uniform distribution were selected to detect 1 chemically mutated elite strain,52 Macadamia hybrid elite strains and 13 parents by PCR amplification and capillary electrophoresis.Finally,7 pairs of SSR markers were used for subsequent data collection,of which one pairs of markers had a low data deletion rate in 66 materials.7 parents were selected as reference varieties,and two groups of parallelexperiments with 7 parents were set up.The establishment of SSR molecular marker database of 53 superior plants and population can provide technical support for the protection of its breeding rights.
