森林草莓FvWRKY46基因的克隆及表达分析

摘要 WRKY转录因子在调控植物生长发育方面有重要作用,但是对于其参与草莓果实品质的影响尚不明确。本研究以森林草莓‘Hawaii4’为材料,克隆转录因子Fv WRKY46并进行生物信息学分析,利用烟草叶片瞬时转化法对FvWRKY46进行亚细胞定位,用RT-qPCR分析FvWRKY46在草莓不同时期和不同组织中的相对表达量。结果表明:FvWRKY46基因全长1 107 bp,共编码368个氨基酸,从森林草莓‘Hawaii4’中克隆到FvWRKY46基因,进行亚细胞定位得知FvWRKY46分布在细胞核中。RT-qPCR分析结果显示FvWRKY46在植物体内各个器官组织均有表达,但在果实中表达量最高,且表达量随着时间的延长在果实成熟期达到最大值。本研究表明,FvWRKY46基因可能参与调控草莓果实成熟的过程。 WRKY transcription factors play important roles in regulating plant growth and development, but its effect on fruit quality remains unclear in strawberry. In this study, the transcription factor Fv WRKY46 was cloned from forest strawberry ’Hawaii4’ and bioinformatics analysis was performed. FvWRKY46 was subcellular localization using tobacco leaf transient transformation method, and the relative expression level of FvWRKY46 in different stages and tissues of strawberry was analyzed by RT-qPCR. The results showed that the total length of FvWRKY46 gene was 1 107 bp, encoding 368 amino acids. FvWRKY46 gene was cloned from strawberry ’Hawaii4’, and the subcellular localization results showed that FvWRKY46 was distributed in the nucleus. RT-qPCR analysis showed that it was expressed in all organs and tissues of plants, but the expression level was the highest in fruits,and the expression level reached the maximum at fruit ripening stage with the extension of time. This study suggested that the FvWRKY46 gene may be involved in regulating the ripening process of strawberry fruit.
