非小细胞肺癌组织中TP酶激活蛋白SH3功能结合蛋白2的表达及临床意义

Expression and clinical significance of TPase-activated protein SH3 functional binding protein 2 in non-small cell lung cancer tissues

目的检测非小细胞肺癌(NSCLC)组织和细胞中TP酶激活蛋白SH3功能结合蛋白2(G3BP2)的表达情况,分析G3BP2对NSCLC细胞增殖、侵袭和凋亡的影响。方法收集NSCLC患者120例,腺癌47例,鳞状细胞癌73例。免疫组织化学检测腺癌、鳞状细胞癌组织中G3BP2的表达情况,分析G3BP2表达与患者临床病理特征的关系。蛋白免疫印迹实验和qRT-PCR检测SPC-A-1、16HBE细胞中G3BP2的表达。对NCI-H226、SPC-A-1细胞转染si-NC、si-G3BP2质粒,检测敲低G3BP2对细胞增殖、侵袭和凋亡的影响。结果腺癌组织和鳞状细胞癌组织中G3BP2的阳性表达率无统计学差异(P>0.05),G3BP2阳性与TNM分期、淋巴结转移有关(P<0.05)。NCI-H226和SPC-A-1细胞中G3BP2蛋白及mRNA的表达高于16HBE细胞。敲低G3BP2可明显抑制NCI-H226、SPC-A-1细胞的增殖和侵袭并增强细胞凋亡。结论G3BP2在NSCLC组织中的阳性表达与TNM分期、淋巴结转移有关,敲低G3BP2的表达可显著抑制NSCLC的进展。

Objective To detect the expression of tumor suppressor TPase-activated protein SH3 functional binding protein 2(G3BP2)in tissues and cells of non-small cell lung cancer(NSCLC),and to analyze the effect of G3BP2 on proliferation,invasion and apoptosis of NSCLC cells.Methods A total of 120 patients with NSCLC,47 with adenocarcinoma and 73 with squamous cell carcinoma were collected.The expression of G3BP2 in adenocarcinoma and squamous cell carcinoma was detected by immunohistochemistry,and the relationship between G3BP2 expression and clinicopathological characteristics of patients was analyzed.The expression of G3BP2 in SPC-A-1 and 16HBE cells were detected by western blot and qRT-PCR.NCI-H226 and SPC-A-1 cells were transfected with si-NC and si-G3BP2 plasmids,and the effect of knockdown of G3BP2 on cell proliferation,invasion and apoptosis were detected.Results There was no significant difference in the positive expression rate of G3BP2 between adenocarcinoma tissue and squamous cell carcinoma tissue(P>0.05).The positive expression of G3BP2 was related to TNM stage and lymph node metastasis(P<0.05).The expression of G3BP2 protein and mRNA in NCI-H226 and SPC-A-1 cells was higher than that in 16HBE cells.Knockdown of G3BP2 significantly inhibited the proliferation and invasion of NCI-H226 and SPC-A-1 cells and enhanced apoptosis.Conclusion The positive expression of G3BP2 in NSCLC tissue is related to TNM stage and lymph node metastasis,and knockdown of G3BP2 expression can significantly inhibit the progression of NSCLC.
