抗双链DNA抗体室内质控品的制备及性能评价

摘要 目的:探讨抗双链DNA(dsDNA)抗体室内质控品的制备方法,并对其精密度和稳定性的质控性能进行评价。方法:对本实验室不同浓度抗dsDNA抗体血清标本进行选择性收集,依据目标靶值将所收集到的血清按比例调配,经灭活、离心、分装、标记、冻存等步骤制备成高值、低值质控品。各测定20管上述高值、低值质控品,确定两个质控品的初始靶值,评估其批内精密度。随后将其作为室内质控品应用于抗dsDNA抗体的日常检测中,将1年内的质控数据按季度分为4组,比较各组均值与初始靶值的差异,并绘制Levey-Jennings质控图,监测质控品的在控状态,评估质控品的稳定性。结果:自制高值、低值抗dsDNA抗体质控品批内变异系数(CV%)分别为5.148%和5.510%;1年内批间CV%分别为5.450%和7.960%,均在试剂厂家提供的CV%可接受范围内(批内≤10%,批间≤15%),Levey-Jennings质控图少有出现失控现象。结论:自制抗dsDNA抗体室内质控品的精密度较高,制备过程简便,在-20℃保存条件下自制质控品的稳定期至少1年,在目前尚未有优质商品质控供用的情况下,适合各级医院实验室推广应用。
