针刺腧穴“降压方”对自发性高血压大鼠ACE-AngⅡ-AT1R轴及Profilin-1的影响 被引量:1

摘要 目的研究针刺腧穴“降压方”对自发性高血压大鼠(SHR)的降压效应及对ACE-AngⅡ-AT1R轴、Profilin-1的影响,探讨针刺降压的作用机制。方法按随机数字表法将30只SHR均分为模型组、针刺组,另以15只Wistar Kyoto Rats(WKR)作为正常对照组。针刺组取双侧人迎穴、曲池穴、足三里穴进行针刺,模型组随意在大鼠躯干部位进行针刺,对照组不针刺,其他操作相同。每周连续干预6 d休息1 d,连续4周,共24次。采用大鼠无创血压测量系统测量血压,并记录一般行为变化,采用ELISA法检测大鼠血清中血管紧张素转化酶(ACE)、血管紧张素Ⅱ(AngⅡ)含量,采用免疫组化法(IHC)检测主动脉中血管紧张素Ⅱ受体1(AT1R)、前纤维蛋白-1(Profilin-1)蛋白表达的水平。结果针刺组第2周开始收缩压、舒张压持续降低,模型组收缩压、舒张压逐渐升高,对照组血压未见明显变化;针刺组较模型组一般行为得到改善。实验4周后针刺组血清中ACE、AngⅡ较模型组显著降低,AT1R、Profilin-1表达水平降低。结论针刺腧穴“降压方”对SHR降压效应明确,其机制可能是通过降低SHR血清中ACE、AngⅡ含量,影响SHR主动脉组织中的AT1R和Profilin-1表达,进而抑制局部肾素-血管紧张素-醛固酮系统(RAAS)的活性起到降低血压的作用。 Objective:To study the anti-hypertensive effect of acupuncture with"Jiangya Fang"on spontaneously hypertensive rats(SHR)and its effects on ACE-AngⅡ-AT1R axis and profilin-1,and to explore the mechanism of acupuncture antihypertensive.Methods:30 SHRs were randomly divided into model group and acupuncture group,and another 15 WKRs were used as normal control group.In the acupuncture group,both sides of"Renying","Quchi"and"Zusanli"were used for acupuncture intervention.In the model group,acupuncture was randomly performed on the trunk of rats,while in the control group,no acupuncture was performed,and other operations were the same as those in the other two groups.The experiment was intervened for 6 days a week,rested for 1 day,and intervened for 4 weeks and a total of 24 times.The blood pressure of rats was measured by noninvasive blood pressure measurement system,and the general behavior changes were recorded.The contents of ACE and AngⅡin rat serum were detected by ELISA,and the expression levels of AT1R and profilin-1 protein in aorta were detected by IHC.Results:The systolic and diastolic blood pressure in the acupuncture group decreased continuously at the second week of acupuncture intervention,and the systolic and diastolic blood pressure in the model group increased gradually,but there was no significant change in the blood pressure in the control group.The general behavior of acupuncture group was improved compared with model group.After continuous acupuncture intervention for 4 weeks,the serum ACE and AngⅡin the acupuncture group were significantly lower than those in the SHR model group,and the expression levels of AT1R and profilin-1 were lower.Conclusion:The anti-hyper-tensive effect of acupuncture with"Jiangya Fang"on SHRs is definite.Its mechanism may be related to reducingthe content of ACE and AngⅡin SHR serum,affecting the expression of AT1R and profilin-1 in SHR aorta,andthen inhibiting the activity of local renin-angiotensin-aldosterone system to reduce blood pressure.
