基于全长转录组数据的尼泊尔黄堇WD40基因家族鉴定及进化分析

摘要 WD40(WD40 domain-containing proteins)转录因子家族广泛参与调节植物生长、发育、次生代谢物积累和环境适应等过程并发挥着重要作用。本研究利用生物信息学手段对25个WD40家族成员的理化特性、启动子顺式作用元件、保守基序、结构域、进化关系和蛋白三级结构进行了分析。结果表明:尼泊尔黄堇中多数WD40蛋白为酸性,且大多为不稳定蛋白,25个WD40蛋白的脂肪族系数均小于100,为亲水性蛋白,并且二级结构以无规则卷曲和延伸链为主,亚细胞定位发现大多在细胞核中。尼泊尔黄堇WD40蛋白具有共同的保守结构域WD40且数量在1~7之间变化。对家族各成员启动子序列中包含的顺式作用元件进行了分析,发现该基因家族含有多种胁迫及植物激素相关的顺式作用元件。进化树分析将尼泊尔黄堇WD40蛋白家族成员分为7个亚家族(Ⅰ~Ⅶ),推测不同类型的WD40蛋白功能存在差异。本研究首次鉴定了尼泊尔黄堇WD40转录因子家族,并揭示了结构特征,这将为深入解析尼泊尔黄堇WD40转录因子在其响应逆境胁迫和其次生代谢产物积累方面的生物学功能提供重要参考。 WD40(WD40 domain-containing proteins), as a transcription factor family, plays an important role and is widely involved in regulating plant growth, development, secondary metabolite accumulation and environmental adaptation. In this study, the physical and chemical properties, promoter cis-acting elements, conserved motifs,structural domains, evolutionary relationships and protein tertiary structure of 25 WD40 family members were analyzed by bioinformatics methods. The results showed that most of the WD40 proteins in Corydalis hendersonii Hemsl. were acidic, and most of them were unstable proteins. The aliphatic coefficients of 25 WD40 proteins were all less than 100, and they were hydrophilic proteins. The secondary structure was dominated by random coils and extended chains, and the subcellular localization was mostly found in the nucleus. The WD40 protein of Corydalis hendersonii Hemsl. has a common conserved domain WD40 and the number varies from 1 to 7. The cis-acting elements contained in the promoter sequences of each member of the gene family were analyzed and it was found that the gene family contained multiple cis-acting elements related to stress and plant hormones. The phylogenetic tree analysis divided the members of the WD40 protein family of Corydalis hendersonii Hemsl. into 7 subfamilies(Ⅰ~Ⅶ), and it is speculated that the functions of different types of WD40 proteins are different. In this study, the WD40 transcription factor family of Corydalis hendersonii Hemsl. WD40 has been identified for the first time and the structural features have been revealed. This will provide important references for further studies on the biological functions of WD40 transcription factor in response to stress and the accumulation of secondary metabolites in Corydalis hendersonii Hemsl 3Hems.
