摘要
白芨,兰科植物,多用于药用和观赏,市场需求较大。为了解决白芨资源短缺、繁殖周期长等问题。以白芨种子为外植体,在1/2MS+2.0 mg/L 6-BA+1.0 mg/LNAA液体培养基中悬浮培养20 d后,以白芨膨大的种子为材料,利用植物组织培养技术,研究不同植物生长调节剂对白芨愈伤组织诱导、增殖和分化的影响,建立白芨愈伤组织诱导及植株再生体系。结果表明,白芨愈伤组织诱导和增殖的较优培养基为MS+1 mg/L6-BA+2 mg/L 2,4-D,诱导率为93.24%,增殖倍数可达到4.17倍。白芨愈伤组织分化的较优培养基为MS+0.1 mg/L 6-BA+0.1 mg/L NAA,分化率为36.50%,将不同的愈伤组织进行分类,分化率最高可达到77.15%。生根的较优培养基为MS+1 mg/L 6-BA+0.8 mg/L NAA,生根率为86.28%。通过愈伤组织诱导建立再生体系,为白芨遗传转化和人工繁育提供了依据。
Bletilla striata, a member of the orchid family, it is mainily used for medicine and ornamental, and has a large market demand. In order to solve the problem of B. striata resource shortage and long reproduction cycle.B. striata seeds were selected the explant for the initial culture in 1/2MS with 2.0 mg/L 6-BA and 1.0 mg/L NAA liquid medium. After 20 days suspension culture, swollen seeds of B. striata were used as the materials to study the effects of the plant growth regulators on induction, proliferation, and differentiation of B. striata callus by plant tissue culture techniques. To establish the callus induction and plant regeneration system of B. striata. As the study shows, the better culture medium for the callus induction and proliferation is MS +1 mg/L 6-BA+2 mg/L 2,4-D.Callus induction rate was 93.24%. The proliferation coefficient was 4.17. MS+0.1 mg/L 6-BA+0.1 mg/L NAA is the optimal medium for the differentiation of callus. The differentiation rate was 36.50%. Classify the different callus, the differentiation rate was up to 77.15%. The optimal medium for rooting of B. striata is MS+1 mg/L 6-BA+0.8 mg/L NAA. The rooting rate was 86.28%. Plant regeneration system of B. striata was established by callus induction, which provided an basis for genetic transformation and artificial propagation.
