不同发根农杆菌菌株对番茄毛状根诱导率及阳性率的影响 被引量:1

Effects of Agrobacterium rhizogenes Strains on Induction Rate and Positive Rate of Hairy Roots

摘要 为了提高番茄毛状根的发根效率,试验以野生型番茄品种Ailsa Craig(AC)为材料,分别使用发根农杆菌菌株Ar Qual、MSU440、C58C1、K599对其进行番茄毛状根诱导,以研究4种不同的发根农杆菌菌株对番茄转基因毛状根诱导率及阳性率的影响。结果表明:发根农杆菌菌株的不同对番茄毛状根诱导率及阳性率均有影响。其中,Ar Qual的发根诱导率最高,约为90.4%,毛状根阳性率约为60.7%;其次是MSU440,发根诱导率约为47.3%,毛状根阳性率约为52.3%;K599、C58C1的发根诱导率较低,分别约为2.5%、0.6%,毛状根阳性率分别约为65%、33.3%。通过该研究找到了更适宜番茄毛状根转化的菌株Ar Qual,这对优化番茄毛状根转化体系,快速获得番茄转基因根系植物材料以开展基因功能、南方根结线虫等方面的研究工作有重大意义。 In order to improve the inducing efficiency of tomato hairy roots with Agrobacterium rhizogenes, the wild-type tomato variety Ailsa Craig(AC) was used as material to induce tomato hairy roots with four kinds of Agrobacterium rhizogenes respectively, such as Ar Qual, MSU440, C58C1 and K599. The results showed that the different strains of Agrobacterium rhizogenes had distinct effects on the induction rate and positive rate of tomato hairy roots. Ar Qual showed the highest induction rate of hairy roots(90.4%) and positive rate of hairy roots(60.7%).Secondly,the induction rate of MSU440 was 47.3% and the positive rate was 52.3%. The induction rates of K599and C58C1 were about 2.5% and 0.6%, and the positive rates were about 65% and 33.3%, respectively. Ar Qual strain was proved to be more suitable for the transformation system in tomato hairy root, which lay the foundation of the rapid acquisition of tomato transgenic root materials for gene function and the study of Meloidogyne incognita.
