基于流式细胞术和基因组Survey的花榈木基因组大小及特征分析 被引量:2

摘要 花榈木是重要的材用、药用及园林绿化树种,国家二级重点保护野生植物。由于基因组数据缺乏,导致花榈木基因资源开发和分子机制研究相对滞后。本研究通过流式细胞术和基因组Survey分析方法对花榈木基因组大小进行估测,获得花榈木基因组重复系列比例、基因组杂合度以及GC含量等信息。经流式细胞术测定,得知花榈木基因组大小约为3 Gb。对花榈木基因组测序数据采用K-mer 17进行分析,得到花榈木基因组大小为3.01 Gb,与前者所得结果基本一致;杂合率为1.04%,重复序列比例为82.23%。采用K-mer 41进行组装,得到的contig N50为656 bp,总长为1 343 713 339 bp,scaffold N50为762 bp,总长为1 364 689 951 bp。根据各信息分析,得知花榈木基因组为高复杂基因组,本研究结果可以为后续基因组组装相应策略提供参考。 Ormosia henryi is an important timber, medicinal and landscaping tree species, belonged to the secondary protected plants. Due to the lack of genome data, the development of its gene resources and the research of its molecular mechanism lag behind. In this study, the genomic size of O. henryi was estimated by flow cytometry and genome survey analysis, and the proportion of genomic repeat series, genomic heterozygosity and GC content of O. henryi were obtained. Flow cytometry showed that the genome size of O. henryi was about 3 Gb. Using K-mer17 for survey analysis, the genomic size of O. henryi was 3.01 Gb, which was basically consistent with the results of the former. The heterozygosity rate was 1.04%, and the proportion of repeat sequences was82.23%. Used K-mer 41 for assembly, the obtained contig N50 was 656 bp, with a total length of 1 343 713 339 bp, and the scaffold N50 was 762 bp, with a total length of 1 364 689 951 bp. The results of this study can provide references for the corresponding strategies of subsequent genome assembly.
